基于血脂水平代谢探讨苏木乙酸乙酯提取物对T2DM大鼠模型胰岛素抵抗及胰腺损伤的干预作用 被引量:1

摘要 目的:基于血脂水平代谢探讨苏木乙酸乙酯提取物对T2DM大鼠模型胰岛素抵抗及胰腺损伤的干预作用。方法:随机选取10只大鼠为空白组,其余大鼠采用高脂高糖饲料+一次性腹腔注射STZ75mg/kg建立T2DM动物模型。各组大鼠分别给予相应药物灌胃,连续给药6周,检测各组大鼠空腹动态血糖值,末次给药2h后,各组大鼠腹主动脉取血。采用酶联免疫Elisa法检测各组大鼠血清中糖化血清蛋白(GSP)、胰岛素(INS)、胰岛素自身抗体(IAA)、胰高血糖素样肽-1(GLP-1)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)水平。取全血检测血液流变学相关指标,取胰腺观察胰腺病理损伤。结果:苏木乙酸乙酯提取物可有效降低模型动物血清中空腹动态血糖值、GSP、IAA、TC、TG、LDL-C含量及血液流变学相关指标,升高INS、GLP-1含量,有效修复受损胰腺,与模型组比较,差异具有统计学意义(P<0.05或P<0.01)。结论:苏木乙酸乙酯提取物可有效降低T2DM血糖值,改善体内血脂紊乱及胰岛素抵抗,增强胰岛素敏改性,修复受损胰腺,达到良好降糖效果。 Objective:To investigate the effect of ethyl acetate extract of Sappan on insulin resistance and pancreatic injury in T2 DM rats.Methods:10 animals were randomly selected as the blank group.The other animals were fed with high-fat and high sugar diet plus STZ 75 mg/kg intraperitoneal injection to establish T2 DM animal model.The animals in each group were given corresponding drugs by gavage for 6 weeks.The fasting blood glucose was detected.2 hours after the last administration,blood samples were taken from the abdominal aorta of each group.The serum glycosylated serum protein(GSP),insulin(INS),insulin autoantibody(IAA),glucagon like peptide-1(GLP-1),total cholesterol(TC)and glycyrrhizin like peptide(GLP-1)were detected by enzyme-linked immunosorbent assay(ELISA),Triglycerides(TG),high density lipoprotein(HDL-C)and low density lipoprotein(LDL-C)were detected.The whole blood was taken to detect the hemorheology related indexes.The pancreas was taken to observe the pathological injury of pancreas.Results:Compared with the model group,the ethyl acetate extract of Sappan could effectively reduce the fasting blood glucose,GSP,IAA,TC,TG,LDL-C content and hemorheology related indexes,increase the contents of ins and GLP-1,and effectively repair the damaged pancreas,with statistical significance(P<0.05 or P<0.01).Conclusion:The ethyl acetate extract of Sappan can effectively reduce the blood glucose value of T2DM,improve the blood lipid disorder and insulin resistance in vivo,enhance the sensitivity modification of insulin,repair the damaged pancreas,and achieve good hypoglycemic effect.
