金莲花总黄酮对T2DM大鼠血清炎症因子、脂代谢及氧化应激的影响 被引量:5

Effects of Total Flavonoids of Trollius Chinensis on Serum Inflammatory Factor,Lipid Metabolism and Oxidative Stress in T2DM Rats

摘要 目的:探讨金莲花总黄酮(TFFTC)对T2DM大鼠血清炎症因子、脂代谢及氧化应激干预作用。方法:随机选择50只Wistar大鼠作为空白组(K),其余用高脂高糖饮食喂养4周。腹腔内注射STZ70mg/kg建立T2DM动物模型。将模型动物随机分为模型组(M)、盐酸二甲双胍组(Y)、金莲花总黄酮高剂量组(TFFTC-G)和金莲花总黄酮低剂量组(TFFTC-D)。每组动物通过灌胃法给予相应药物治疗6周,每2周测量1次空腹血糖,腹主动脉取血测定血清糖化血清蛋白(GSP)、胰岛素(INS)、胰岛素自身抗体(IAA)、血脂四项(总胆固醇-TC、甘油三酯-TG、高密度脂蛋白-HDL-C、低密度脂蛋白-LDL-C)、超氧化物气化酶(SOD)、丙二醛(MDA),总抗氧化能力(T-AOC)、肿瘤坏死因子(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)等指标影响。结果:与模型组比较,盐酸二甲双胍组及金莲花总黄酮高、低剂量组可不同程度降低模型大鼠血清动态血糖值,降低血清中GSP、IAA、TC、TG、LDL-C、TNF-α、IL-1β、IL-6及MDA含量,升高模型动物血清中INS、SOD、T-AOC含量,差异具有统计学意义(P<0.05或P<0.01)。结论:金莲花总黄酮可有效降低T2DM血清血糖水平,降低血清炎症因子表达及氧化应激反应发生,调节血脂功能紊乱,达到良好降糖效果。 Objective:To investigate the effects of total flavonoids of Trollius chinensis on serum inflammatory factors,lipid metabolism and oxidative stress in T2 DM rats.Methods:50 Wistar rats were randomly selected as blank group(k).The rest were fed with high fat and high sugar diet for 4 weeks.The animal model of T2 DM was established by intraperitoneal injection of STZ 70 mg/kg.The model animals were randomly divided into model group(M),metformin hydrochloride group(Y),TFFTC-G and TFFTC-D.Fasting blood glucose was measured every two weeks.Blood samples were taken from abdominal aorta to determine serum glycosylated serum protein(GSP),insulin(INS),insulin autoantibody(IAA),four items of blood lipids(total cholesterol TC,triglyceride TG,high density lipoprotein-HDL-C,low density lipoprotein-LDL-C),superoxide dismutase(SOD),superoxide dismutase(LDL-C)Malondialdehyde(MDA),total antioxidant capacity(T-AOC),tumor necrosis factor(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)and other indicators.Results:Compared with the model group,the metformin group and the high and low flavonoid groups of the total flavonoids of the Flos Flos could reduce the serum glucose level of the model animals in different degrees,and reduce the contents of GSP,IAA,TC,TG,LDL-C,TNF-α,IL-1β,IL-6 and MDA in the serum,and increase the levels of INS,SOD,and MDA in the serum of the model animals,with statistical significance(P<0.05 or P<0.01).Conclusion:Total flavonoids of Trollius chinensis can effectively reduce the level of serum glucose in T2 DM,reduce the expression of serum inflammatory factors and the occurrence of oxidative stress,regulate dyslipidemia,and achieve good hypoglycemic effect.
