摘要
目的:基于网络药理学分析淫羊藿-三七配伍治疗前列腺癌的潜在靶点及相关机制。方法:借助TCMSP及TCMID数据库检索淫羊藿、三七的药物活性成分及靶点信息,GeneCards及OMIM数据库检索前列腺癌的作用靶点。利用Cytoscape软件构建"药物-成分-靶点-疾病"网络图,通过STRING及CentiScape进行PPI蛋白互作分析,同时对药物-疾病关键靶点进行GO和KEGG富集分析。结果:共检索淫羊藿-三七的有效活性成分24个,治疗前列腺癌靶点基因64个,筛选出64个核心靶点。GO富集分析中生物学过程集中在泛素化、核受体功能、转录相关功能、激素受体功能、细胞凋亡等;KEGG富集分析提示淫羊藿-三七治疗前列腺癌主要涉及细胞凋亡、肿瘤抑制信号通路(p53)、内分泌抵抗、磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)、肿瘤坏死因子(TNF)等信号通路。结论:该研究初步预测了淫羊藿-三七配伍治疗前列腺癌的关键靶点以及相关信号通路,提示其可能通过促进细胞凋亡、抑制肿瘤细胞增殖、降低内分泌抵抗、杀伤肿瘤细胞以及抑制肿瘤血管生成等途径发挥抗肿瘤作用,为进一步深入研究提供了新思路。
Objective:To analyze the potential targets and related mechanisms of Epimedium-Panax notoginseng compatibility in the treatment of prostate cancer based on network pharmacology.Methods:The TCMSP and TCMID databases were used to retrieve the active ingredients and target genes of Epimedium-Panax notoginseng,and the targets of prostate cancer were retrieved from the GeneCards and OMIM databases.Visual analysis of“drug-component-target-disease”network with Cytoscape.With the help of STRING and CentiScape,PPI core genes were screened,and GO function enrichment analysis and KEGG signal pathway enrichment analysis were performed on key targets of drug-disease.Results:A total of 24 active ingredients compatible with Epimedium-Panax notoginseng were retrieved and 64 target genes compatible with Epimedium-Panax notoginseng were used to treat prostate cancer.Based on this,“drug-component-target-Disease”network visualization analysis,screened out 64 PPI core genes.The enrichment of GO function prompts the biological process and function of Epimedium-Panax notoginseng to focus on ubiquitination,nuclear receptor function,transcription-related function,hormone receptor function,and apoptosis;KEGG pathway enrichment The analysis suggests that the treatment of Epimedium-Panax notoginseng on prostate cancer mainly involves apoptosis,tumor suppressor signal pathway(p53),endocrine resistance,phosphatidylinositol 3-kinase/protein kinase B(PI3 K/AKT),tumor Signaling pathways such as necrosis factor(TNF).Conclusion:The medicine has the characteristics of multi-pathway and multi-target effect on EpimediumPanax notoginseng on the treatment of prostate cancer.This study initially analyzed the key potential targets of the drug’s compatibility with Epimedium-Panax notoginseng and related intracellular signal transduction pathways,suggesting that it can promote apoptosis,inhibit tumor cell proliferation,reduce endocrine resistance,and kill The tumor cells and the inhibition of tumor angiogenesis play an anti-tumor role,providing a certain reference and reference for the further research.
