牛樟叶水提物对急性酒精性肝损伤的保护作用及其机制研究

Protective Effect and Mechanism of Aqueous Extracts of Cinnamomum Kanehirae Hayata Leaves on Acute Alcohol-induced Liver Injury in Mice

目的:观察牛樟叶水提物(AECL)对急性酒精性肝损伤的保护作用及其机制。方法:取健康KM小鼠,随机分为正常组,模型组,联苯双酯组(0.2g/kg),AECL高(20g/kg)、中(10g/kg)、低(5/kg)剂量组,除模型组外,其他各组给予药物灌胃,每天2次,连续10d。末次给药0.5h后,除正常组给予蒸馏水外,其余各组一次灌胃56°白酒12mL/kg,禁食不禁水12h后,取肝脏制备成10%组织匀浆和分离血清,检测ALT、AST、SOD、GSH、MDA、TG含量。结果:与模型组比较,AECL高、中剂量组小鼠血清中ALT、AST活性显著降低,GSH水平和SOD活性显著升高;高剂量组血清中MDA和TG水平显著降低。高、中剂量肝组织中GSH水平显著升高;高剂量肝组织中SOD活性显著增强,MDA水平降低。结论:牛樟叶水提物具有明显改善急性酒精性肝损伤的作用,其作用机制可能与其抗氧化作用有关。

Objective:To observe the Aqueous Extracts of Cinnamomum Kanehirae Hayata Leaves,AECL on acute alcoholic liver injury protection mechanism..Methods:Healthy KM mice were randomly divided into normal group,model group,biphenyl diesters group(0.2 g/kg),high AECL group(20 g/kg),medium group(10 g/kg),and low(5 g/kg)groups.In addition to model group,other groups were given drug gavage twice a day for 10 d.After 0.5 hof the last administration,the normal group was given distilled water,and the other groups were given 12 mL/kg of 56°white wine.After fasting for 12 h,the liver was taken to prepare 10%tissue homogenate and isolated serum,and the contents of ALT,AST,SOD,GSH,MDA and TG were detected.Results:Compared with the model group,the serum ALT and AST activities in the high-dose and medium-dose AECL groups were significantly decreased,and the serum GSH levels and SOD activities were significantly increased.Serum MDA and TG levels were significantly reduced in the highdose group.The level of GSH in high and medium dose liver tissue was significantly increased.SOD activity was significantly increased and MDA level decreased in high dose liver tissue.Conclusion:The Aqueous Extracts of Cinnamomum Kanehirae Hayata Leaves can obviously improve acute alcoholic liver injury,and its mechanism may be related to its antioxidant effect.
