活血散结中药复方含药血浆对bFGF干预下兔RPE细胞Bax、Bcl-2表达的影响

Effects of Plasma Containing Invigorating Blood and Dissipating Masses Chinese Medicine Compound on Bax and Bc1-2 Protein Expression of Rabbit RPE Cell Under bFGF Intervention

目的:探索活血散结中药复方含药血浆对碱性成纤维细胞生长因子(bFGF)干预下兔视网膜色素上皮(RPE)细胞Bax、Bcl-2蛋白表达的影响。方法:选用对数生长期兔RPE细胞,分为正常血浆组、bFGF组、bFGF+BGJ398组、bFGF+含药血浆组,Western blotting法检测各组兔RPE细胞Bcl-2、Bax蛋白表达情况,观察活血散结中药复方含药血浆作用后RPE细胞Bcl-2、Bax蛋白在兔RPE细胞中的表达。结果:bFGF+含药血浆组与bFGF比较,Bax蛋白表达明显增强,Bcl-2蛋白表达明显抑制,差别均有显著统计学意义(P<0.01)。结论:活血散结中药复方含药血浆作用于bFGF干预下所致增生性玻璃体视网膜病变(PVR)的细胞模型,可促进RPE细胞Bax蛋白的表达,同时抑制Bcl-2蛋白的表达,使Bax/Bcl-2构成比升高。

Objective:To explore the effects of plasma containing invigorating blood and dissipating masses Chinese medicine on Bax and Bc1-2 protein expression of rabbit RPE cell under bFGF intervention.Methods:Select the rabbit RPE cells in log phase.The experiment objects were divided into the normal plasma group,the bFGF group,the bFGF+BGJ398 group,the bFGF+drug-containing plasma group.Western Blotting method was used to detect the Bcl-2,Bax protein expression condition in rabbit RPE cells and the Bcl-2,Bax protein expression condition in rabbit RPE cells after being acted by invigorating blood and dissipating masses Chinese medicine compound was observed.Results:Compared with bFGF group,Bax protein expression of bFGF+drug-containing plasma group was strengthened evidently,and Bcl-2 expression inhabitation was lightened,and the difference was statistically significant(P<0.01).Conclusion:Plasma containing invigorating blood and dissipating masses Chinese medicine acts on PVR cell model caused by bFGF intervention,promotes Bax protein expression and inhibits Bcl-2 protein expression,reducing the constituent ratio of Bax/Bcl-2.
