基于网络药理学探讨麻黄-杏仁配伍增效减毒的科学内涵 被引量:7

摘要 目的:基于网络药理学方法,探讨麻黄-杏仁配伍增效减毒的分子生物学机制。方法:通过数据库获取麻黄、杏仁的活性成分及靶点,结合疾病数据库获取支气管哮喘的相关靶点,利用Cytoscape软件分别构建两药"成分-靶点"网络与蛋白相互作用网络,使用ClueGO插件分别对麻黄、杏仁作用靶点进行GO与KEGG分析,通过对比两药作用靶点、生物过程、作用通路的异同,探讨麻黄、杏仁配伍增效减毒的科学内涵。结果:麻黄可作用于JUN、TP53、IL6、FOS、IL10等靶点抑制炎性免疫反应,对气道平滑肌的作用较弱;杏仁可调节CD38、NOS2、ESR1、COL-1、CHRM2蛋白,扩张支气管,抑制气道重塑。同时杏仁可调节血压与心肌的兴奋性,缓解麻黄的不良反应,麻黄可通过调节缺氧反应、细胞凋亡等生物过程,降低杏仁的毒副作用。结论:麻黄与杏仁作用靶点互补,配伍使用即可增强彼此药理作用,又可抑制彼此的毒副作用。网络药理学研究为中药配伍研究提供了新的思路。 Objective:To explore the effect of ephedrae herba combined with amygdalus communis Vas on the efficacy-enhancing and toxicity-reducing using network pharmacology techniques.Methods:According to the database to obtain the components of ephedrae herba and amygdalus communis vas and their target.Acquired the bronchial asthma-related targets through the disease databases,The Cytoscape software was used to construct"compound-target network"and"protein-protein interaction"network.The ClueGO software was used for GO and KEGG analysis of potential genes.By comparison of target,GO analysis and KEGG pathway of ephedrae herba and amygdalus communis vas in the treatment of bronchial asthma,the scientific connotation of effect-enhancing and toxicity-reducing functions of ephedrae herba combined with amygdalus communis was discussed.Results:Ephedrae herba can regulate JUN,TP53,IL6,FOS,IL10 to inhibit inflammatory reaction,but the effect on airway smooth muscle is weak.Amygdalus communis vas dilate bronchus and inhibit airway remodeling by regulating CD38,NOS2,ESR1,COL-1 and CHRM2 proteins.At the same time,amygdalus communis vas can regulate blood pressure and myocardial excitability,alleviate the adverse reactions of ephedrae herba.Ephedrae herba can reduce the toxic and side effects of amygdalus communis vas by regulating the hypoxia and apoptosis.Conclusion:The targets of ephedrae herba and amygdalus communis vas are complementary,which has the effect of synergism and toxicity reduction.Network pharmacology technology provides a new idea for the study of compatibility of traditional Chinese medicine.
