黄精地龙配伍对自然衰老小鼠学习记忆及海马神经凋亡的影响 被引量:4

摘要 目的:探讨黄精地龙配伍对自然衰老小鼠学习记忆及海马神经凋亡的影响。方法:用2、4、8g·kg-1·d-1剂量的黄精地龙配伍浓缩提取液灌胃18月龄昆明小鼠4周,然后采用跳台试验、旷场试验分别观察该配伍对衰老小鼠学习记忆能力、焦虑程度、自发活动与探索行为能力的影响,采用尼氏染色、免疫组化分别检测该配伍对衰老小鼠海马CA1及CA3区神经元数量、Bcl-2与Caspase-3表达的变化。结果:黄精地龙配伍可明显增强衰老小鼠学习记忆能力,提高自发活动与探索行为能力,降低焦虑程度,增加海马各区神经元数量,并能升高海马各区Bcl-2表达及降低Caspase-3表达。结论:黄精地龙配伍可改善衰老小鼠学习记忆功能,提高其自发活动与探索行为能力并降低焦虑程度,这与该配伍通过抗海马神经凋亡机制发挥神经保护作用有关。 Objective:To explore the effect of compatibility of Huangjing and Dilong(HD)on learning and memory and apoptosis of hippocampal nerve in natural aging mice.Methods:The 18 month old Kunming mice were given 2,4,8 g·kg-1·d-1 HD by gavage for 4 weeks.Then,the effects of HD on learning and memory ability,anxiety level,spontaneous activity and exploration behavior ability of aging mice were observed by platform jumping test and open field test respectively,the changes of neuron number,Bcl-2 and Caspase-3 expression in hippocampal CA1 and CA3 of aging mice were tested by Nissl staining and immunohistochemistry respectively.Results:HD could significantly enhance the learning and memory ability of aging mice,improve the ability of spontaneous activity and exploration behavior,reduce the level of anxiety,increase the number of neurons of the hippocampus,and increase the expression of Bcl-2 and Caspase-3 of the hippocampus.Conclusion:HD could improve the learning and memory function of aging mice,improve their spontaneous activity and explore behavior ability,and reduce the level of anxiety,which is related to its neuroprotection through the mechanism of anti hippocampal nerve apoptosis.
