摘要
目的:建立高效液相色谱法测定中风回语颗粒中大黄酚的含量。方法:采用Dimonsil C18色谱柱(5μm,250mm×4.6mm),流动相为0.1%磷酸水-甲醇(20∶80),检测波长为254nm,流速为1mL·min-1,柱温30℃。结果:大黄酚在10.05~160.90μg·mL-1范围内线性关系良好(r=1.0000),精密度、稳定性试验RSD均<0.50%,重复性试验RSD<2.30%,平均加样回收率(n=6)为98.16%,RSD=1.17%。结论:该方法重复性好、准确度高,可用于中风回语颗粒中大黄酚的含量测定。
Objective:To establish a high performance liquid chromatography(HPLC)method for determination of chrysophanol in Zhongfeng Huiyu Granules.Methods:The separation was performed on a Dimonsil C18 column(5μm,250 mm×4.6 mm)with the mobile phase of 0.1%phosphoric acid-methanol(20∶80),the detection wavelength was 254 nm,the flow rate was 1 mL·min-1,the column temperature was 30℃.Results:The response of chrysophanol was liner in the range of 10.05~160.90μg·mL-1,the RSD of the precision and stability test were lower than 0.50%,the RSDof the repeatability test were lower than 2.30%,the average recovery(n=6)was 98.16%,RSD was 1.17%.Conclusion:The established method was reliable,reproducible,which can be used for the determination of chrysophanol in Zhongfeng Huiyu Granules.
