盐炙杜仲对维甲酸诱导的骨质疏松大鼠骨生长因子及骨代谢的影响 被引量:2

摘要 目的:探究盐炙杜仲对维甲酸诱导的骨质疏松大鼠骨生长因子的影响。方法:选取60只雄性SD大鼠,随机分为6组:空白组、模型组、阳性对照组、杜仲低剂量组(DL)、杜仲中剂量组(DM)、杜仲高剂量组(DH),10只/组。空白组0.5%CMC-Na灌胃,其余组给予同等剂量的维甲酸持续灌胃2周。造模成功后,各组大鼠分别给予相应的药物灌胃,模型组和空白组给予等体积的0.5%CMC-Na,阳性对照组给予骨疏康颗粒,杜仲低、中、高剂量组分别给予1.5、3、6g/kg剂量杜仲,每天1次,共4周后,检测6组骨痂骨密度(BMD)和矿物含量(BMC)、雌二醇(E2)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)、骨生长因子(BMP-2、VEGF、TGF-β、IGF-1)水平。结果:与空白组相比,模型组大鼠BMD、BMC、E2、BMP-2、VEGF、TGF-β、IGF-1水平明显下降(P<0.05),TNF-α、IL-6水平显著升高(P<0.05)。与模型组相比,阳性对照组、杜仲中、高剂量组大鼠BMD、BMC、E2、BMP-2、VEGF、TGF-β、IGF-1水平有不同程度上升(P<0.05),TNF-α、IL-6水平有明显降低趋势(P<0.05)。结论:实验表明,中、高剂量杜仲可通过调节骨生长因子水平,提高相关骨生长因子表达,提高骨密度,对维甲酸所致的大鼠骨质疏松有较好的防治作用。 Objective:To observe the effects of salt-processed Eucommia ulmoides on bone growth factor and metabolism biochemical indexes in retinoic acid-induced osteoporosis rats.Methods:60 male SD rats were randomly divided into six groups:blank control group,model group,positive control group(Gushukang granules),Eucommia ulmoides low/medium/high dose group(DL/DM/DH).The blank group was given 0.5%CMC-Na solution,the other groups were given retinoic acid to establish the osteoporosis model for 2 weeks.After successful modeling,the rats in each group were given corresponding drugs by gavage,the model group and blank group were given equal volume of 0.5%CMC-Na,the positive control group was given Gushukang granules,and the low,medium and high dose Eucommia ulmoides groups were given 1.5,3 and 6 g/kg Eucommia ulmoides respectively,once a day.After 4 weeks,detection of six groups of rats E2、BMP-2、VEGF、TGF-β、IGF-1、TNF-α、IL-6 and the change of BMD and BMC.Results:Compared with the blank group,BMD、BMC、E2、BMP-2、VEGF、TGF-β、IGF-1 levels of the model group were significantly decreased(P<0.05),and the levels of TNF-α、IL-6 were significantly increased(P<0.05).Compared with the model group,the BMD、BMC、E2、BMP-2、VEGF、TGF-β、IGF-1 levels of rats in the positive control group and the middle and high dose groups of Eucommia ulmoides significantly improved(P<0.05),and the levels of TNF-α、IL-6 showed a significant decrease(P<0.05).Conclusion:The experiment shows that middle and high doses of Eucommia ulmoides has a good effect on osteoporosis induced by retinoic acid in rats.
