小儿解表颗粒对发热大鼠的解热作用及机制研究 被引量:3

摘要 目的:观察小儿解表颗粒对LPS、2,4-二硝基酚和干酵母3种不同模型诱导大鼠发热的解热作用,探讨不同浓度小儿解表颗粒对2,4-二硝基酚发热模型的解热机制。方法:筛选出体温稳定大鼠预防给药3d后,分别注射LPS、2,4-二硝基酚和干酵母进行造模,造模后测量不同时间段大鼠的体温,筛选出小儿解表颗粒解热效果最佳的发热模型及最佳解热时间。对2,4-二硝基酚发热模型的大鼠在体温达到最高点麻醉取血和下丘脑,酶联免疫吸附试验(ELISA)技术检测下丘脑中cAMP、NO、PGE2和血清中IL-1β、IL-6、TNF-α、IFN-γ、GHRL、β-EP的表达。结果:小儿解表颗粒对2,4-二硝基酚诱导大鼠的发热模型解热效果最好,且最佳解热时间在造模后20 min左右。2,4-二硝基酚模型组大鼠下丘脑cAMP、NO、PGE2和血清IL-1β、IL-6、TNF-α、IFN-γ、GHRL、β-EP的表达均增加(P<0.05),经过小儿解表颗粒治疗后,治疗组与模型组相比,各指标的表达均降低(P<0.05)。结论:小儿解表颗粒对2,4-二硝基酚诱导大鼠发热具有显著解热效果,其作用机制与其能影响发热大鼠外周介导发热的细胞因子、中枢升温介质及相关炎症因子含量的表达有关。 Objective:To observe the antipyretic effect of Xiao Er Jie Biao Granule on the fever induced by LPS,2,4-dinitrophenol and dry yeast in rats,and to explore the antipyretic mechanism of different concentrations of Xiao Er Jie Biao Granule on the 2,4-dinitrophenol fever model.Methods:The rats with stable temperature were screened,and then the drug was administered for 3 days.Then LPS,2,4-dinitrophenol and dry yeast were injected respectively.The body temperature of rats in different time periods was measured after modeling.Finally,the best fever model and the best antipyretic time were obtained.Rats with 2,4-dinitrophenol fever model were anesthetized to take blood and hypothalamus when their body temperature reached the highest point.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of cAMP,NO and PGE2 in the hypothalamus and IL-1β,IL-6,TNF-α,IFN-γ,GHRL andβ-EP in serum.Results:Xiao Er Jie Biao Granule had the best antipyretic effect on the fever model induced by 2,4-dinitrophenol in rats,and the best antipyretic time was about 20 minutes after modeling.The expression of cAMP,NO,PGE2 in the hypothalamus and IL-1β,IL-6,TNF-α,IFN-γ,GHRL andβ-EP in serum of 2,4-dinitrophenol model group increased(P<0.05),after treatment with Xiao Er Jie Biao Granule,the expression of each index was lower in the treatment group than in the model group(P<0.05).Conclusion:Xiao Er Jie Biao Granule has significant antipyretic effect on 2,4-dinitrophenol induced fever in rats.Its mechanism of action is related to the expression of cytokines,central warming medium and related inflammatory factors in peripheral blood-mediated fever in rats with fever.
