基于HPLC-ELSD指纹图谱和含量测定的薏苡仁药材质量评价研究

Quality Evaluation of Semen coicis Based on HPLC-ELSD Fingerprint and Determination

目的:建立薏苡仁药材HPLC-ELSD指纹图谱,并测定甘油三油酸酯的含量,为薏苡仁药材质量评价提供依据。方法:采用Merck Purospher?STAR RP-18e(250mm×4.6mm,5μm)色谱柱,以乙腈-二氯甲烷(65∶35)为流动相进行等度洗脱,建立薏苡仁药材HPLC-ELSD指纹图谱,并进行相似度评价、聚类分析及主成分分析,同时测定甘油三油酸酯的含量。结果:18批薏苡仁药材指纹图谱有8个共有峰,相似度均大于0.9,聚类分析及主成分分析将18批药材分为3类,与相似度评价结果一致;不同产地的薏苡仁甘油三油酸酯的含量范围为0.70%~1.91%,以云南产的1批薏苡仁药材含量最高,其他产地的薏苡仁药材甘油三油酸酯的含量较稳定。结论:HPLC-ELSD指纹图谱结合定量检测可以有效评价薏苡仁药材的产品质量。

Objective:To provide a basis for quality assessment of semen coicis by establishing HPLC-ELSD fingerprint of semen coicis and determining the content of glycerol trioleate.Methods:Chromatographic column of Merck Purospher?STAR RP-18 e(250×4.6 mm,5μm)was used with acetonitrile,dichloromethane(65∶35)as mobile phase for equi degree elution.The fingerprint of semen coicis was established and evaluated by similarity calculation,cluster analysis and principal component analysis.At the same time,the content of glycerol trioleate was determined.Results:There were 8 common peaks in the fingerprint of 18 batches of semen coicis and the similarity was greater than 0.9.Cluster analysis and principal component analysis divided 18 batches of semen coicis into 3 categories,which were consistent with the similarity evaluation results.The content of triglyceride in coix seed from different producing areas ranged from 0.70%to 1.91%.The content of one batch of Coix seed from Yunnan was the highest,and the content of triglyceride in coix seed from other producing areas was sta=ble.Conclusion:HPLC-ELSD fingerprint combined with quantitative detection can be effectively used for the quality evaluation of Coix seed.
