苦木药材中总生物碱的含量测定及聚类分析和判别分析 被引量:1

Determinination of the Content of Total Alkaloids in the Picrasma quassioides (D.Don) Benn and Its Quality Evaluation

摘要 目的:测定不同产地苦木药材中总生物碱的含量,优化现有苦木药材标准中总生物碱含量测定方法,评价苦木药材的质量。方法:采用紫外分光光度法,于318 nm处测定吸光度,以苦木酮为对照品,测定苦木药材中总生物碱的含量。结果:苦木酮的线性范围为0.004~0.02 mg/mL(r=0.9999),线性关系良好;加样回收率在94.51%~103.94%之间,平均回收率(n=9)为100.22%(RSD=2.8%),准确度良好;精密度RSD小于3.0%,精密度良好。通过聚类分析和偏最小二乘法判别分析(PLS-DA),结果表明用药部位对总生物碱含量影响最大,产地次之。结论:不同产地不同部位苦木药材中总生物碱含量差异较大,总生物碱含量与产地和部位呈一定的相关性。 Objective:To determine the content of total alkaloids in the Picrasma quassioides(D.Don)Benn from different areas and different medicinal parts,to optimize the existing method for determination of its total alkaloids,and to evaluate the quality of it.Methods:UV spectrophotometry was used to detect the concent of total alkaloid at 318 nm by using the nigakinone as reference substance.Results:The linear range of nigakinone was 0.004~0.02 mg/mL,r=0.9999,the recovery was 94.51%~103.94%,average recovery was 100.22%,RDS=2.8%,the RSD of precision was less than 0.3%,the results shows that this method with good linear relation,recovery,precision and accuracy.The results show that position of Picrasma quassioides(D.Don)Benn has the most important influence on total alkaloid and it is followed by producing area,analysis by cluster analysis and PLS-DA.Conclusion:The difference of the content of total alkaloids in the Picrasma quassioides(D.Don)Benn from different areas and different medicinal parts was statistically significant,There is correlation between contents of total alkaloids and localities and parts.
