黄芪多糖对肺纤维化大鼠mTOR、p-S6、α-SMA的影响 被引量:9

Effect of Astragalus Polysaccharide on mTOR、p-S6、α-SMA in rats with Pulmonary Fibrosis

摘要 目的:观察黄芪多糖(APS)对博莱霉素(BLM)致肺纤维化大鼠哺乳动物雷帕霉素靶蛋白(mTOR)信号通路的影响。方法:实验动物随机分为6组,采用BLM气管注入法复制肺纤维化模型,造模后分别给予生理盐水、APS、雷帕霉素(RAPA)灌胃,连续给药28天后提取样本。采用HE和Masson染色观察大鼠肺组织病理学改变,免疫组化法观察大鼠肺组织中p-S6、α-SMA、mTOR的表达。结果:HE和Masson染色结果显示,APS可降低肺纤维化大鼠的肺泡炎积分和肺组织纤维化积分;免疫组化结果显示,APS和雷帕霉素可抑制肺纤维化大鼠肺组织中p-S6、α-SMA、mTOR的表达。结论:APS可能通过抑制mTOR及其下游p-S6的表达,影响α-SMA生成,干预肺纤维化的发生。 Objective:Observe the effects of Astragalus Polysaccharide to Pulmonary Fibrosis rats cause by BLM in mTOR signaling pathways.Methods:The experimental animals were divided into six groups.The pulmonary fibrosis model was copied by bleomycin tracheal injection method for all the groups,except blank control group.When the rat models copied,use normal saline,APS and RAPA gavage administration keep 28 days.Observe the pathological changes by HE and Masson staining;observe the express about mTOR,p-S6,α-SMA by IHC.Results:HE staining shows that Astragalus Polysaccharide can reduce the alveolar inflammation integral in rats,compare to the BLM model group.Masson staining shows that APS and RAPA can reduce the fibrosis integral of lung tissue in rats,compare to the BLM model group.The result display for IHC,the express about p-S6,α-SMA,mTOR were inhibited by APS and RAPA.Conclusion:The results shows that APS may inhibit the mTOR and p-S6 to reduce the expression aboutα-SMA which is the characteristics of muscle fibroblasts to intervene the progress in Pulmonary Fibrosis.
