甲亢型肾阴虚的模型复制研究

摘要 目的:研究甲状腺素复制甲亢型肾阴虚大鼠模型的效果。方法:根据随机数字表将雄性SD大鼠分为正常组和肾阴虚模型组,以160 mg·kg^(-1)甲状腺素片灌胃21 d复制甲亢型肾阴虚模型。日常观察大鼠的饮食量、饮水量、体重、肛温等一般指标,采集大鼠甲状腺、胸腺、肾上腺等脏器以及血液、肾脏样本;计算脏器指数,采用酶联免疫吸附法测定血清中的三碘甲状腺原氨酸(T3),四碘甲状腺原氨酸(T4),雌二醇(E2),睾酮(T),环磷酸腺苷(cAMP),环磷酸鸟苷(cGMP);通过HE染色观察肾脏病理变化。结果:与正常组比较,甲亢型肾阴虚模型组较活跃、不易抓取、暴躁、毛发脱落严重,饮水量增加,肛温升高,痛阈值降低,尿量增多,血糖升高;血清中的T3、T4、E2/T、cAMP/cGMP升高;肾小管病变严重,结构紊乱,管腔扩大形成空泡;肾小管上皮细胞萎缩、减少或者脱落,染色质缩小;肾小球结构紊乱。结论:甲状腺素片复制肾阴虚模型效果良好,可应用于实际肾阴虚模型的复制中。 Objective:To study the effect of thyroxine on the replication of hyperthyroidism kidney yin deficiency rat model.Methods:According to the random number table,male SD rats were divided into normal group and kidney yin deficiency model group.The hyperthyroidism kidney yin deficiency model was replicated by intragastric administration of 160mg·kg^(-1)thyroxine tablets for 21 days.General indicators such as dietary amount,drinking amount,weight and rectal temperature of rats were observed ordinarinessly,and samples of thyroids,thymus,adrenal glands,blood and kidneys were collected.The visceral index were calculated,and triiodothyronine(T3),tetraiodothyronine(T4),estradiol(E2),testosterone(T),cyclic adenosine monophosphate(cAMP)and cyclic guanosine monophosphate(cGMP)in serum were detected by enzyme linked immunosorbent assay.Renal pathological changes were observed by HE staining.Results:As compared with the normal group,more active,difficulty to grasp,petulance,falling off of hair seriously.Drinking amount,rectal temperature,amount of urine and blood glucose were increased.Pain threshold was decreased.T3、T4、E2/T、cAMP/cGMP were increased.Renal tubular lesions were serious,and its structure was disordered.The lumen expanded to form vacuole.Renal tubular epithelial cells atrophied,reduced or fallen off.Its chromatin minified.The structure of glomeruli was disordered.Conclusion:The use of thyroxine tablets to replicate the kidney yin deficiency model is effective,which can be applied to the replication of kidney yin deficiency model in reality.
