摘要
目的:测定美花石斛提取物的多糖含量浓度,探究不同干燥方法下美花石斛提取物的多糖含量的不同及建立美花石斛药材定性鉴别的薄层色谱方法,以期为美花石斛质量标准的建立提供参考。方法:运用薄层色谱法鉴别其多糖,并用硫酸-苯酚法测定美花石斛提取物中多糖的含量。结果:薄层色谱的斑点显示清晰,斑点圆型规整且重现性好,含量测定的平均回收率为100.01%(n=6),方法精密度RSD为3.7%(n=6)。综合考虑目前在加热沸腾干燥法下测定时,多糖的含量(33.42%)相对较高。结论:该方法操作简便、准确,目前可用于美花石斛薄层色谱定性鉴别。
Objective:Determine the polysaccharide content concentration of Dendrobium loddigesii extract,explore the differences in polysaccharide content of Dendrobium loddigesii extract under different drying methods,and establish a thin layer chromatography method for qualitative identification of Dendrobium loddigesii,in order to provide reference for the establishment of quality standards for Dendrobium loddigesii.Methods:The polysaccharide was identified by Thin-layer chromatography,and the content of polysaccharide in the extract of Dendrobium loddigesii was determined by sulfuric acid phenol method.Result:The spots in thin-layer chromatography were clearly displayed,with regular circular patterns and good reproducibility.The average recovery rate for content determination was 100.01%(n=6),and the RSD of the method precision was 3.7%(n=6).Considering the current situation,the content of polysaccharides(33.42%)is relatively high when measured using the heating boiling drying method.Conclusion:This method is simple and accurate,and can currently be used for qualitative identification of Dendrobium loddigesii by thin layer chromatography.
