摘要
目的:通过评价对α-葡萄糖苷酶和二肽基肽酶-4(DPP-4)的活性抑制作用,研究石菖蒲根、茎和叶的降糖作用。方法:采用系统溶剂提取法制备石菖蒲根、茎和叶的不同极性提取物,分别进行α-葡萄糖苷酶和DPP-4酶活测试试验,计算各提取物对α-葡萄糖苷酶和DPP-4的抑制率和半数抑制浓度(IC_(50))。结果:石菖蒲的正己烷、二氯甲烷、乙酸乙酯、正丁醇、甲醇和水提物II均对α-葡萄糖苷酶有抑制作用,其中茎的乙酸乙酯提取物抑制活性最强,其IC_(50)为6.83μg/mL;其正己烷、二氯甲烷、乙酸乙酯、正丁醇、甲醇、水提物I、水提物II均对DPP-4有抑制作用,其中根的乙酸乙酯提取物抑制活性最强,其IC_(50)为155.37μg/mL。结论:石菖蒲提取物能一定程度地抑制α-葡萄糖苷酶和DPP-4的活性。
Objective:To study the hypoglycemic activity of roots,stems and leaves of Acorus tatarinowii by evaluating the inhibition of α-glucosidase and DPP-4 activity.Methods:Different polar extracts of Acorus tatarinowii root,stem and leaf were extracted by systematic solvent extraction method.The α-glucosidase and DPP-4 activity test were conducted with all extracts,and the inhibition rate and half inhibition concentration(IC_(50))of α-glucosidase and DPP-4 were calculated.Results:In α-glucosidase study,n-hexane,dichloromethane,n-butanol,ethyl acetate,methanol,and aqueous extract II of Acorus tatarinowii showed inhibitory effect,among which the ethyl acetate extract of stem has the strongest inhibitory activity,IC_(50) was 6.83μg/mL.The extracts of n-hexane,dichloromethane,ethyl acetate,n-butanol,methanol,aqueous extract I and aqueous extract II preformed the inhibitory effects on DPP-4,among which the ethyl acetate extract of root has the strongest inhibitory activity,IC_(50) was 155.37μg/mL.Conclusion:Acorus tatarinowii extract can inhibit the activity ofα-glucosidase and DPP-4.
