艾纳香提取物对α-葡萄糖苷酶及DPP-4的抑制作用研究

摘要 目的:研究天然植物艾纳香根、茎、叶、花四个部位对α-葡萄糖苷酶和DPP-Ⅳ的抑制作用,筛选出艾纳香对α-葡萄糖苷酶和DPP-Ⅳ有抑制作用的提取部位。方法:采用系统溶剂提取法对艾纳香的四个植物部位进行提取。通过体外酶活测定实验,评估提取物对α-葡萄糖苷酶和DPP-Ⅳ酶的抑制能力。结果:艾纳香根、茎、叶、花的氯仿、正丁醇、甲醇提取物对α-葡萄糖苷酶有抑制作用;其中,艾纳香花的氯仿、甲醇提取物抑制作用最强,IC_(50)分别是7μg/mL和9μg/mL。艾纳香根、茎、叶、花的正丁醇提取物和叶、花的甲醇提取物对DPP-Ⅳ有抑制作用;其中,艾纳香花的正丁醇、甲醇提取物的抑制作用最强,IC_(50)分别是93μg/mL和67μg/mL。艾纳香提取物对α-葡萄糖苷酶和DPP-Ⅳ的抑制作用呈现浓度依赖关系。结论:艾纳香提取物对α-葡萄糖苷酶和DPP-Ⅳ均有不同程度的抑制作用,其降糖活性成分主要集中于氯仿、甲醇和正丁醇提取物中,,可进一步从这些提取物中寻找潜在的降糖活性成分。本实验为研究艾纳香提供实验基础,为艾纳香降血糖研究实验依据。 Objective:To investigate the inhibitory effects of a-glucosidase and DPP-IV from the root,stem,leaf,and flower of Blumea balsamifera(L.)DC.(BB).To screen the active extracts with inhibitory effects onα-glucosidase and DPP-4 from BB.Methods:Systematic solvent extraction method was used to extract the different polar extracts of BB parts.The effect of extracts on a-glucosidase and DPP-IV were evaluated by extracellular enzyme activity test.Results:The extracts of chloroform,nbutanol and methanol from root,stem,leaf and flower had inhibitory effect onα-glucosidase,while chloroform and methanol extract of flower showed the strongest inhibition,IC_(50)were 7μg/mL and 9μg/mL,respectively.The n-butanol extracts of root,stem,leaf and flower and the methanol extracts of leaf and flower had inhibitory effects on DPP-Ⅳ,n-butanol and methanol extracts of flower performed the highest inhibitory effect,IC_(50)were 93μg/mL and 67μg/mL,respectively.The inhibitory effects of the extract onα-glucosidase and DPP-Ⅳwere concentration dependent.Conclusion:The extracts of BB have different inhibitory effects onα-glucosidase and DPP-Ⅳ.The hypoglycemic active components in BB are mainly concentrated in chloroform,methanol and n-butanol extracts,and the methanol extract of BB flower presented the stronger inhibition onα-glucosidase and DPP-Ⅳthan other parts.These extracts can further find potential hypoglycemic active ingredient.
