摘要
目的:为了探寻最适合青钱柳新鲜叶片基因组DNA提取的方法。方法:本研究以青钱柳新鲜叶片为材料,以5种不同的基因组提取方法提取青钱柳基因组DNA,并将提取到的基因组DNA经超微量分光光度计、凝胶电泳检测、聚合酶链式反应(PCR)检测,找出最适合提取青钱柳叶片基因组DNA的方法。结果:综合考虑,试剂盒法是最适合用于提取青钱柳DNA的方法,且能满足实验中对总DNA的要求,使得实验能够获得的DNA稳定,且提取效率高。结论:本研究为青钱柳基因组高质量DNA的提取方法选择提供了参考,为后续进行SCoT分子标记实验提供科学依据。
Objective:To explore the best method for genomic DNA extraction from fresh leaves of Cyclocarya paliurus.Methods:In this study,fresh leaves of Cyclocarya paliurus were used as materials to extract genomic DNA from Cyclocarya paliurus with five different methods of genome extraction.The extracted genomic DNA was detected by ultramicro spectrophotometer,gel electrophoresis and polymerase chain reaction(PCR)to find the most suitable method for extracting genomic DNA from Cyclocarya paliurus leaves.Results:Comprehensively,the kit method is the most suitable method for extracting the DNA of Cyclocarya paliurus,and can meet the requirements for total DNA in the experiment,making the DNA obtained in the experiment stable and efficient.Conclusion:This study provides a reference for the selection of high-quality DNA extraction methods of Cyclocarya paliurus genome,and provides a scientific basis for the subsequent SCoT molecular marker experiment.
