Simplicillium lanosoniveum菌株分离鉴定及斜面培养基配方优化

Isolation and Identification of Fungus Simplicillium lanosoniveum and the Optimization of Its Agar Slant Culture-Medium Formulation

分离并鉴定了采自长春净月潭国家森林公园的1株真菌—Simplicillium lanosoniveum,优化了斜面培养基配方,通过单因素和正交试验考察不同培养基质对S.lanosoniveum菌丝生长的影响,筛选最适的碳源、氮源和微量元素,并进行了验证试验。结果表明:在28℃培养温度下,最优培养基配方为麦芽糖5 g/L,酵母粉2.5 g/L,KH;PO;1.5 g/L,马铃薯200 g/L,琼脂粉20 g/L,p H自然,为生防菌剂及抗生素的进一步研发奠定基础。

Simplicillium lanosoniveum,a fungus from Changchun Jingyuetan National Forest Park,was isolated and identified.The optimization of agar slant culture-medium formulation of the strain was investigated by single factor and orthogonal test.The effects of different culture medium on the mycelial growth of S.lanosoniveum were studied,and the optimal carbon,nitrogen and trace elements were selected,then the optimization and verification experiments were carried out.The results showed that the optimal medium was maltose concentration 5 g/L,yeast powder concentration 2.5 g/L,KH;PO;concentration 1.5 g/L,potato concentration 200 g/L,agar powder concentration 20 g/L,with a natural pH value at 28℃,which laid the foundation for further research and the development of biological control agent and antibiotics.
