摘要
[目的]探讨组蛋白赖氨酸甲基转移酶(SETD1B)对肺癌细胞增殖和凋亡的影响.[方法]对肺癌A549细胞进行SETD1B siRNA转染,实验分为正常组、siNS组和siSETD1B组,均用浓度为0.1μmol/L处理,转染48 h,采用细胞集落形成实验检测细胞增殖能力,利用流式细胞术检测细胞周期及细胞凋亡情况.[结果]与siNS组和正常组比较,siSETD1B组细胞增殖能力显著降低,细胞周期受到阻滞,细胞凋亡率明显升高,相比较差异均有统计学意义(P<0.05).[结论]沉默SETD1B可抑制肺癌细胞增殖,阻滞细胞周期并诱导细胞凋亡.
OBJECTIVE To explore the effects of histone lysine methyltransferase(SETD1 B)on the proliferation and apoptosis of lung cancer cells.METHODS Lung cancer A549 cells were transfected with SETD1 B siRNA,and the experiment was divided into normal group,siNS group and siSETD1 B group.All cells were treated with concentration of 0.1μmol/L for 48 h.Cell proliferation was detected by cell colony formation assay,and cell cycle and apoptosis were detected by flow cytometry.RESULTS Compared with the siNS group and the normal group,cell proliferation ability of the siSETD1 B group was significantly decreased,cell cycle was arrested,and cell apoptosis rate was significantly increased,and the differences were statistically significant(P<0.05).CONCLUSION SETD1 B silencing can inhibit the proliferation of lung cancer cells,block cell cycle and induce apoptosis.
