朝医太阴中风散对脑缺血再灌注大鼠脑组织的保护作用及其机制

摘要 [目的]探讨朝医太阴中风散对脑缺血再灌注大鼠脑组织的保护作用及其机制.[方法]利用线拴法制作大鼠脑缺血模型,随机分为假手术组、模型对照组、阳性对照组、太阴中风散低、高剂量组.假手术组及模型对照组分别给予生理盐水10 mL/kg,太阴中风散低、高剂量组分别给予太阴中风散10,20 g/kg,阳性对照组给予尼莫地平1 mg/kg,每日1次,连续灌胃给药15 d.利用水迷宫实验检测空间记忆能力,采用HE染色法观察脑组织形态学改变,采用ELISA法检测SOD,MDA水平.[结果]与假手术组比较,模型对照组逃避潜伏期明显延长,穿越站台次数明显减少(P<0.05);与模型对照组比较,太阴中风散高剂量组和阳性对照组逃避潜伏期明显缩短,穿越站台次数明显增多(P<0.05);与模型对照组比较,太阴中风散低、高剂量组神经细胞变性、坏死、脱失均减轻,血清SOD水平均明显升高(P<0.01),MDA水平均明显降低(P<0.05).[结论]朝医太阴中风散对脑缺血再灌注大鼠脑组织具有保护作用,其机制认为可能与增强机体抗氧化性有关系. OBJECTIVE To explore the protective effects and mechanism of traditional Korean medicine Taiyin apoplexy powder on brain tissue of cerebral ischemia reperfusion rats.METHODS Line tether method was used to make cerebral ischemia rat model.Rats were randomly divided into sham operation group,model control group,positive control group,low and high dose of Taiyin apoplexy powder group.The rats in the sham operation group and the model control group were given normal saline 10 mL/kg,the rats in the Taiyin apoplexy Powder low and high dose groups were given Taiyin apoplexy Powder 10 g/kg and 20 g/kg respectively,the rats in the positive control group were given nimodipine 1 mg/kg by continuous gavage once a day for 15 days.The spatial memory ability was detected by morris water maze test,the morphological changes of brain tissue were observed by HE staining,the levels of SOD and MDA were detected by ELISA.RESULTS Compared with the sham operation group,the escape latency of the model control group was significantly prolonged,and the number of crossing platform was significantly reduced(P<0.05).Compared with the model control group,the escape latency of the high dose group and the positive control group was significantly shortened,and the number of crossing platform was significantly increased(P<0.05).Compared with the model control group,the degeneration,necrosis and loss of nerve cells of low and high dose groups of Taiyin apoplexy powder were all reduced,serum SOD level increased significantly(P<0.01),and serum MDA level decreased significantly(P<0.05).CONCLUSION Traditional Korean medicine Taiyin apoplexy powder had protective effects on the brain tissue of cerebral ischemia reperfusion rats,the mechanism was thought to be related to the enhancement of antioxygenation.
