摘要
[目的]探讨DOCK8蛋白在特应性皮炎(AD)调节性T细胞中的作用.[方法]取NC/Nga小鼠建立AD模型,利用AutoMACS仪器从NC/Nga小鼠脾脏细胞中分离调节性T细胞,用促肾上腺皮质激素释放激素(CRH)处理后观察DOCK8表达变化;处理DOCK8 siRNA后观察调节性T细胞的功能性改变.[结果]CRH处理后AD调节性T细胞中DOCK8表达水平明显降低(P<0.01).Real-time PCR及ELISA检测结果显示,调节性T细胞DOCK8表达量降低后细胞因子IL-10和TGF-β表达水平明显降低(P<0.05,P<0.01,P<0.001).[结论]DOCK8蛋白在调节性T细胞中起重要作用,调节性T细胞中DOCK8的表达水平提高可能有助于减轻AD症状.
OBJECTIVE To detect the role of DOCK8 in the regulatory T cells(Tregs).METHODS NC/Nga mice were used to make AD model.Spleens of mice were mashed using syringe plunger to make cell suspension,and Tregs were sorted using AutoMACS.After treating corticotropin-releasing hormone(CRH),DOCK8 expression was detected.After treating DOCK8 siRNA,function of Tregs was detected.RESULTS DOCK8 was downregulated in the CRH-treated AD Tregs(P<0.01).Using real-time PCR and ELISA,I found that inhibitory cytokines TGF-βand IL-10 were deceased after treating with DOCK8 siRNA compared to control group(P<0.05,P<0.01,P<0.001).CONCLUSION DOCK8 plays an important role in Tregs which may contribute to alleviating stress induced aggravation of AD symptom.
