摘要
研究以纤维素酶提取无籽刺梨多糖的最佳条件,并探讨无籽刺梨多糖体外抗氧化活性。以无籽刺梨多糖得率为响应值,在单因素试验的基础上,利用Box-Behnken响应面分析法确定最佳提取工艺条件,并采用1,1-二苯基-2-三硝基苯肼(DPPH)和羟基自由基清除能力体系评价无籽刺梨多糖体外抗氧化活性。无籽刺梨多糖最佳提取工艺条件下的酶用量为3196.53 U/g、p H值为5.18、提取时间为82.46 min和提取温度48.58℃,此条件下模型预测多糖得率为16.85%,无籽刺梨多糖对DPPH和羟基自由基清除能力的IC50值分别为1.75mg/mL和0.83mg/mL。无籽刺梨多糖的纤维素酶法提取工艺方便可行,得到的多糖具有体外抗氧化活性。
To study the optimum extraction conditions of polysaccharide of Rosa sterilis fruit by cellulase and its antioxidant activity in vitro,on the basis of single factor experiment,Box-Behnken response surface analysis was used to determine the optimal extraction conditions,and the in vitro antioxidant activity of polysaccharide of R.sterilis fruit was evaluated by 1,1-diphenyl-2-picrylhydrazyl(DPPH)and hydroxyl radical scavenging ability system.The optimum extraction conditions of polysaccharide of R.sterilis fruit were as follows:enzyme dosage 3196.53 U/g,pH value 5.18,extraction time 82.46 min and extraction temperature 48.58℃.Under these conditions,the polysaccharide extraction rate predicted by the model was 16.85%.IC50 values of DPPH and hydroxyl radical scavenging abilities of polysaccharide of R.sterilis fruit were 1.75 mg/mL and 0.83 mg/mL,respectively.The cellulase extraction of polysaccharide of R.sterilis fruit by cellulase is convenient and feasible,and the obtained polysaccharide has antioxidant activity in vitro.
