八重樱花总黄酮提取工艺响应面法优化及其抗氧化活性研究 被引量:5

Study on Optimization of Extraction Process for Total Flavonoids in Cerasus sp. by Response Surface Methodology and Its Antioxidant Activity

摘要 以干八重樱花为研究对象,通过单因素试验分析及响应面法优化设计,探究乙醇体积分数、温度、液料比对樱花总黄酮提取率的影响,并通过DPPH自由基清除试验、OH自由基清除测试,考察樱花提取液的抗氧化能力。结果显示:樱花总黄酮最佳提取工艺为乙醇体积分数45.7%,温度79.3℃,液料比(体积质量比)50.8 mL/g。在此工艺下,黄酮提取率可达13.306%,通过3组验证试验,结果显示平均黄酮提取率为13.252%,RSD为0.53%,具有良好的准确性。对樱花提取液进行DPPH自由基、OH自由基清除试验,结果显示,以生药量计的樱花提取液对DPPH自由基及OH自由基的IC50值分别为44.1 mg/L和0.80 mg/m L,表明樱花提取液具有良好的抗氧化能力。 Taking dry flowers of Cerasus sp.as the research object,through single-factor experimental analysis and response surface optimization design,the effects of ethanol volume fraction,temperature,liquid-to-material ratio on the extraction rate of total flavonoids in this flower were studied.The antioxidant capacity of its extract was also studied through DPPH and OH free radical scavenging experiments.The results showed that the best extraction process for total flavonoids in Cerasus sp.was ethanol volume fraction 45.7%,temperature 79.3℃,and liquid-to-material ratio 50.8 mL/g.Under this condition,the extraction rate of flavonoids can reach 13.306%.The three sets of verification experiment results showed that the average extraction rate of flavonoids was 13.252%,and the RSD was 0.53%with good accuracy.The results of free radical scavenging experiments showed that the IC50 of Cerasus sp.extract in terms of crude drugs for DPPH free radical and OH radical were 44.1 mg/L and 0.80 mg/mL,respectively,indicating this extract has a good antioxidant capacity.
