黑加仑籽粉对高脂血症小鼠血脂的影响

摘要 目的研究黑加仑籽粉对高脂血症模型小鼠血脂水平的影响。方法雄性昆明种小鼠90只,适应性喂养1 w后,随机抽取15只作为空白对照组给予基础饲料,其余作为建模组用高脂饲料配以高脂饮物喂养。将建模组小鼠随机分为高脂模型组、辛伐他汀阳性对照组(5 mg·kg^(-1)·d^(-1))和黑加仑籽粉低剂量(1.25 g·kg^(-1)·d^(-1))、中剂量(2.5 g·kg^(-1)·d^(-1))、高剂量(5 g·kg^(-1)·d^(-1))组,连续4 w每日灌胃给予各受试物。4 w后,处死动物,分离血清,测定总胆固醇(TC)、甘油三酯(TG)含量;分离肝脏和主动脉,制作石蜡切片,光镜下观察各组小鼠肝脏组织形态变化及基质金属蛋白酶9(MMP-9)在主动脉的表达情况。结果与空白对照组比较,高脂模型组TC、TG明显升高,差异具有统计学意义(P<0.05),表明造模成功;黑加仑籽粉低、中、高剂量组血清TC、TG均较高脂模型组低,差异具有统计学意义(P<0.05),并具有一定的量效关系;黑加仑籽粉低、中、高剂量组可不同程度的缓解小鼠肝细胞肿胀、动脉管壁脂质沉积等病变,以高剂量组作用效果最佳。结论黑加仑籽粉具有降低实验性高脂血症小鼠血脂的功效,并对肝脏及有助于动脉粥样硬化斑块的稳定。 Objective To study the effect of black currant seed powder on blood lipid in hyperlipidermia mice.Methods 90 male mice,fed with regular diet for 1 week,randomly were selected 15 as blank group received basic diet,and others as model group fed with high-fat diet.Randomized them into high fat model group,simvastatin group(5mg·kg^(-1)·d^(-1))and black currant seed powder lowdose(1.25g·kg^(-1)·d^(-1)),middle dose(2.5g·kg^(-1)·d^(-1)),and high dose(5g·kg^(-1)·d^(-1))groups,then with drugs oral delivery for 4 weeks.Finally,collected blood samples and test TC and TG,and separated liver and aorta to make paraffin sections to observe the morphological changes of liver and aorta and the expression level of MMP-9 in aorta by light microscope.Results Compared with blank groups,the levels of serum TC and TG in model group were higher with statistical significance(P<0.05),indicating successful modeling.Blank currant seed powder lowdose,middle dose and high dose group,the serum TC and TG level were lower than hyperlipidemia model group with statistical significance(P<0.05),and there was a dose effect relationship between TC and TG levels in serum of the 3 dose groups black current seed powder.Black currant seed power lowdose,middle dose and high dose group can diversely relieve pathological changes of liver cell edema,thickening of tube wall of aorta,lipid storage and so on.The effect of high dose group was best among these.Conclusion Black currant can reduce the level of blood lipids of experimental mice and the liver and contribute to the stability of atherosclerotic plaque.
