摘要
目的利用分子生物学和生物信息学技术平台对miR-31-3p脑胶质瘤靶基因及相关重要信号通路调节机制,进行分析,为了解miR-31-3p脑胶质瘤调控网络及后续功能验证提供理论基础。方法利用荧光定量PCR技术,检测miR-31-3p在U87细胞中的表达量;合成miR-31-3p模拟物及抑制物,转染至U87细胞,进行链特异性转录组分析,构建miR-31-3p及相关重要信号网络。结果miR-31-3p表达量在U87细胞中显著下降;其潜在靶基因主要涉及细胞增殖、转录及肿瘤等信号通路,这表明miR-31-3p参与生物体多种生化过程,特别是与人类健康重要疾病。
Objective To predict the target genes of miR-31-3p and its regulation mechanism of some important pathways in glioma,and provide the theoretical basis for its experimental verification and regulation study.Methods The expression of MiR-31-3p in U87 cells was detected by quantitative PCR.MiR-31-3p target genes were predicted by using the open single-stranded non-coding small RNA database.MiR-31-3p mimics and inhibitors were synthesized and transfected into U87 cells for chain specific transcriptome analysis and setting up the regulation network of the signal pathway.Results The expression level of MiR-31-3p significantly decreased in U87 cell and MiR-31-3p may negatively regulate some important pathway.Its potential target genes are mainly involved in cell proliferation,transcription and tumor signaling pathways.This suggests that MiR-31-3p is involved in a variety of biological biochemical processes,particularly in important human health diseases.
