摘要
目的探讨EPHX1甲基化水平与癫痫患者卡马西平耐药的关系。方法选取卡马西平耐药癫痫患者25例,卡马西平反应良好的癫痫患者25例,另设置正常对照组25例。使用Cp G搜索器可以预测Cp G岛;亚硫酸氢盐测序PCR(BSP)用于测试EPHX1的甲基化。然后,我们在临床参数和EPHX1甲基化之间进行了统计分析。结果年龄和性别相匹配的CBZ耐药患者,CBZ敏感患者和健康对照者之间无差异。CBZ耐药组的癫痫发作频率,病程和EPHX1甲基化之间存在显着正相关。结论EPHX1启动子的甲基化水平与CBZ耐药性癫痫有关。
Objective To explore the relationship between EPHX1 methylation level and carbamazepine resistance in patients with epilepsy.Methods 25 patients with carbamazepine-resistant epilepsy were selected,25 patients with carbamazepine-responsive epilepsy,and 25 patients with normal control groups.Cp G islands was predicted using the Cp G searcher;bisulfite sequencing PCR(BSP)was used to test the methylation of EPHX1.Then,a statistical analysis between clinical parameters and EPHX1 methylation was performed.Results There were no differences between age and sex-matched CBZ-resistant patients,CBZ-sensitive patients,and healthy controls.There was a significant positive correlation between seizure frequency,disease course and EPHX1 methylation in the CBZ resistance group.Conclusion The methylation level of EPHX1 promoter is related to CBZ-resistant epilepsy.
