庞氏安胎止血汤对热证自然流产大鼠T细胞亚群表达干预的研究

Intervention of Pang’s Antai Zhixue Decoction on expression of T lymphocyte subsets in spontaneous abortion rats with heat syndrome

目的初步探讨庞氏安胎止血汤对热证自然流产模型大鼠血清CD4+、CD8+的表达干预及庞氏安胎止血汤的安胎机制。方法将妊娠SD大鼠随机分为模型组、西药组(地屈孕酮组)、庞氏安胎止血汤高剂量组、庞氏安胎止血汤中剂量组、庞氏安胎汤止血低剂量组和正常组,用附子、干姜、肉桂和米非司酮建立热证自然流产动物模型,末次给药24 h后,腹主动脉取血并分离血清;剖视子宫观察着床数和流产率;酶联免疫法检测大鼠血清CD4+、CD8+表达。结果模型组大鼠流产率、血清CD4+明显升高、CD8+水平显著降低;经治疗后,中药低剂量组、中药中剂量组和西药组大鼠流产率显著降低(P<0.01),中药低剂量组、中药中剂量组、中药高剂量和西药组血清CD4+明显降低,CD8+水平显著升高。结论庞氏安胎止血汤可能通过降低热证自然流产大鼠血清中T淋巴细胞亚群CD4+,升高CD8+,使CD4+/CD8+的平衡向CD8+方向转移,发挥安胎作用。

Objective To investigate the intervention of Pang,Shi An Tai Zhi Xue Decoction on the expression of serum CD4^+and CD8^+in spontaneous abortion model rats and the tocolysis mechanism of Pang,Shi Tuo Zhi Xue Decoction.Methods Pregnancy SD rats were randomly divided into model group,western medicine group(Dihydrated progesterone group),Pang’s Antuo Zhixue Decoction high-dose group,Pang’s Antai Zhixue Decoction middle-dose group,and Pang’s Antuyu Decoction to stop bleeding.In the dose group and the normal group,an animal model of spontaneous abortion with hot syndrome was established with Fuzi,ginger,cinnamon,and mifepristone.After the last 24 hours of dosing,the abdominal aorta was taken and the serum was separated;the uterus was sectioned to observe the number of beds and abortions rate;Enzyme-linked immunosorbent assay to detect the expression of CD4^+and CD8^+in rat serum.Results The abortion rate,serum CD4^+and CD8^+levels in the model group were significantly increased,and the CD8^+level was significantly decreased.After treatment,the low dose group,the middle dose group and the western medicine The abortion rate in the rats was significantly lower(P<0.01).The serum CD4^+levels in the low-dose group,medium-dose group,high-dose Chinese medicine group,and western medicine group significantly decreased,and the CD8^+level increased significantly.Conclusion Pang’s Antai Zhixue Decoction may play a role in tocolysis by decreasing the serum T lymphocyte subsets CD4^+,increasing CD8^+,and shifting the balance of CD4^+/CD8^+to CD8^+in spontaneous abortion rats.
