淫羊藿含药血清对BMSCs中Sirt1mRNA的表达的影响 被引量:5

摘要 目的探讨淫羊藿含药血清和白藜芦醇对慢病毒转染后骨髓基质干细胞Sirt1 mRNA表达的影响。方法将骨髓基质干细胞随机分为空白组、慢病毒载体感染组、慢病毒西药组、慢病毒中药组,应用淫羊藿和白藜芦醇对慢病毒转染的骨髓基质干细胞进行干预。观察各组治疗前后Sirt1mRNA表达的变化。结果分别于干预开始后1d、3d、7d、14d检测,结果发现慢病毒中药组与空白组比较,促进Sirt1表达作用明显,具有显著性差异(P<0.01);与慢病毒载体感染组比较,1d、3d、7d、14d检测,慢病毒中药组促进Sirt1表达作用明显,差异具有显著意义(P<0.01),与慢病毒西药组比较,在1d,3d,14 d检测时,慢病毒中药组具有促进Sirt 1表达作用明显,差异具有显著意义(P<0.01),在7 d检测时,慢病毒中药组促进Sirt1mRNA表达升高差别无统计学意义(P>0.05)。结论淫羊藿促进慢病毒转染后骨髓基质干细胞在成骨分化过程中Sirt1 mRNA的表达,促进成骨细胞生成,达到预防和治疗骨质疏松的目的。 Objective To observe the effects of serum containing Epimedium at different concentrations on osteogenic differentiation of bone marrow stromal stem cells(bone marrow mesenchymal stem cells,BMSCs)and expression of Sirt1(Silent information regulator homolog1)mRNA after lentivirus transfection in castrated rats.Methods The cell were randomly divided into control group,lentivirus vector infection group,lentivirus vector infection combined with middle concentration traditional Chinese medicine group,lentivirus vector infection combined with Resveratrol.The serum containing Epimedium intervention in BMSCs,observed Sirt1 mRNA expression changes.Results After intervened of 1 d,3 d,7 d,14 d.The blank group and infected with lentiviral vector compared with traditional Chinese medicine group.the BMSCs Sirt1 mRNA expression was significantly increased(P<0.01),and lentiviral vector infection combined with Resveratrol,the BMSCs Sirt1 mRNA expression was significantly increased within 1 d,3 d,14 d,(P<0.01),The BMSCs Sirt1 mRNA expression was not significantly increased within 7 d(P>0.05).Conclusion Epimedium medicated serum can promote osteoporosis rats lentivirus transfected MSCsand its expression in Sirt1 mRNA osteogenic differentiation process,thus inhibit the adipogenic differentiation of MSCs,to achieve the prevention and treatment of osteoporosis.
