摘要
目的通过改良纤维素测定方法,找到更加简便测定滇黄精纤维素含量的方法。方法对国标GB/T 5515-2008中纤维素测定仪法进行分步骤条件改良,根据相对标准偏差和测定结果得出针对性强、较简便的测定方法。结果相同管重复3次和不同管同时测定的相对标准偏差分别为0.033和0.038,表明该仪器具有良好的重现性;添加硅藻土有利于提高测定结果的稳定性;脱脂处理对结果无影响,脱脂处理可以忽略不计;碳酸盐去除过程对结果没有影响,可省去处理;加入消泡剂是必要的;氢氧化钠和氢氧化钾在碱消解过程中对测定结果没有差异,因此不做特殊要求。结论经过改良的粗纤维素含量测定方法针对性强,适用于滇黄精粗纤维素含量分析。不同地区滇黄精粗纤维素含量存在一定差异,为发掘优质食用滇黄精资源提供了基础数据。
Objective An improved method to analyze the crude cellulose was studied in order to find a rapid determination way of the crude cellulose content in Polygonatum kingianum.Methods A simple and targeted methods of the standard GB/T 5515-2008 ware got according to the relative standard deviation and the determination results in cellulose conditions after improving cellulose conditions step by step.Results(1)The relative standard deviations of 6 tubes on one time and one tube on three times were 0.038 and 0.033,which indicated that the test instrument was reproducible;(2)adding Celite was helpful to improve the stability of the determination results;(3)The degreasing treatment had no effect on the results,and the degreasing treatment could be negligible;(4)The process of removing carbonate had no effect on the results could be negligible;(5)The addition of defoamers was necessary for the test;(6)The sodium hydroxide and the potassium hydroxide in the alkali digestion process had no difference in the determination of the results.Conclusion The improved method was highly targeted and could be used to determinate the cellulose contents of Polygonatum kingianum.There were some differences in the content of crude cellulose among different Polygonatum kingianum germplasms.It set a basic date to develop high quality edible variety for Polygonatum kingianum.
