基于网络药理学探讨血必净治疗胰腺炎相关机制

Mechanism of Xuebijing in treatment of pancreatitis based on network pharmacology

摘要 目的采用网络药理学方法探讨血必净治疗胰腺炎的相关机制。方法采用TCMSP数据库分析血必净的活性成分及作用靶点,采用Disgenet、OMIM、GeneCards、Phenopedia、Phenolyzer数据库分析胰腺炎相关靶点,获取血必净与胰腺炎的交集靶点。采用CytoNCA软件构建交集靶点网络图,STRING软件绘制PPI网络图,DAVID软件分析KEGG通路和GO富集。结果筛选获得79个血必净活性成分,对应靶点232个,疾病靶点208个,血必净与胰腺炎的交集靶点35个。GO富集分析得到GO条目30个,其中分子功能10个,生物过程条目10个,细胞组成条目10个。KEGG通路分析涉及HIF-α信号通路,PI3K-Akt信号通路,JAK-STAT信号通路等。结论应用网络药理学方法阐明血必净治疗胰腺炎的作用靶点及通路,为血必净治疗胰腺炎提供理论与实验依据。 Objective To explore the mechanism of Xuebijing in treating pancreatitis based on network pharmacology.Methods Active components and corresponding targets of Xuebijing were acquired via the database of Traditional Chinese Medicine Systems Pharmacology Database(TCMSP).The targets associated with severe pancreatitis were analyzed via the databases,including Disgenet,OMIM,GeneCards,Phenopedia and Phenolyzer.The intersection targets were acquired by integrating the Xuebijing targets and severe pancreatitis related targets.The CytoNCA software was applied to analyze the"active component-target"network and STRING software was utilized to drawn protein-protein interaction(PPI)network.KEGG pathway analysis and GO enrichment analysis were carried out via DAVID database.Results 79 active components of Xuebijing and 232 corresponding targets were obtained.In addition,208 targets associated with pancreatitis were found.Our further analysis showed that there were 35 drug and disease intersection targets.Based on GO enrichment analysis,10 biological process terms,10 molecular function terms and 10 cellular component terms were acquired.KEGG pathway was enriched and screened to get 30 signaling pathways,such as HIF-αsignaling pathway,PI3K-Akt signaling pathway,JAK-STAT signaling pathway and so on.Conclusions The targets and pathways of Xuebijing in the treatment of pancreatitis are elucidated by network pharmacology,which provides a theory and experimental basis for the further clarification of Xuebijing in the treatment of pancreatitis.
