淫羊藿苷对脓毒症小鼠急性肺损伤的作用 被引量:1

Effect of icariin on sepsis-induced acute lung injury in mice

摘要 目的探讨淫羊藿苷对脓毒症小鼠急性肺损伤的作用及机制。方法按照随机数字表将8周龄雄性Balb/c小鼠随机分为3组:假手术组、模型组和实验组,每组16只。用盲肠结扎穿刺法建立脓毒症模型。实验组在造模后用淫羊藿苷(30mg/kg,灌胃)干预,假手术组及模型组给予等量0.5%DMSO灌胃。以称重法检测肺湿干重比值;伊文思蓝(EB)法检测肺微血管渗漏;酶联免疫吸附法检测支气管肺泡灌洗液(Bronchoalveolar lavage fluid,BALF)中肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)水平;以比色法检测肺组织丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性;苏木素-伊红(HE)染色观察肺组织病理变化并镜下评分。结果与假手术组比较,模型组肺湿干重比升高(P<0.01),肺组织EB含量增加(P<0.01),BALF中的TNF-α、IL-1β及IL-6水平升高(P<0.01),肺组织MDA含量增加、SOD活性下降(P<0.01),肺组织的损伤评分升高(P<0.01);与模型组比较,实验组肺湿干重比降低(P<0.01),肺组织EB含量降低(P<0.05),BALF中的TNF-α、IL-1β及IL-6水平降低(P<0.01),肺组织MDA含量降低、SOD活性升高(P<0.01),肺组织的损伤评分降低(P<0.01)。结论淫羊藿苷对脓毒症诱导的急性肺损伤具有一定的保护作用,其机制可能与抑制炎症反应与氧化应激、降低微血管渗漏有关。 Objective To explore the effect and mechanism of icariin on acute lung injury in septic mice.Methods Balb/c male mice aged 8 weeks were randomly divided into three groups according by random number table:sham group,model group and experimental group.Sepsis was conducted by cecal ligation and puncture(CLP)and icariin(30 mg/kg,by gavage)was given post-CLP in experimental group.The sham and model group were given the same amount of 0.5%DMSO.The ratio of wet-to-dry weight of lung was measured;pulmonary microvascular leakage was detected by evans blue(EB).The levels of tumor necrosis factor alpha(TNF-α),Interleukin-1β(IL-1β)and interleukin-6(IL-6)in bronchoalveolar lavage fluid(BALF)were detected by enzyme-linked immunosorbent assay.Malondialdehyde(MDA)content and superoxide dismutase(SOD)activity in lung tissue were detected by colorimetry.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of lung tissue and the score was calculated under microscope.Results Compared with the sham group,Lung wet-to-dry weight ratio,the EB content,the levels of TNF-α、IL-1βand IL-6 in the BALF,MDA content in the lung,the lung injury score increased greatly in the CLP group(P<0.01),SOD content in the lung decreased in the CLP group(P<0.01).Compared with the CLP group,Lung wet-to-dry weight ratio,the EB content,the levels of TNF-α、IL-1βand IL-6 in the BALF,MDA content in the lung,the lung injury score decreased obviously in experimental group(P<0.05 or P<0.01),SOD content in the lung increased in experimental group(P<0.01).Conclusions Icariin may ameliorate sepsis-induced acute lung injury in mice by inhibiting inflammatory response,oxidative stress and reducing microvascular leakage.
