摘要
目的研究软枣猕猴桃提取物对H2O2诱导血管内皮细胞损伤的保护作用。方法用过氧化氢(H2O2)诱导血管内皮细胞损伤,采用四甲基偶氮噻唑蓝(MTT)比色法检测软枣猕猴桃提取物对细胞增殖的影响,放射免疫法检测其对内皮素(ET-1)及前列环素(PGI2)释放的影响。结果过氧化氢对血管内皮细胞具有损伤作用,软枣猕猴桃提取物能显著对抗过氧化氢引起的血管内皮细胞损伤,显著抑制乳酸脱氢酶(LDH)渗漏、显著增加PGI2释放及显著减少ET-1释放。结论软枣猕猴桃提取物对过氧化氢诱导的血管内皮损伤具有显著保护作用。
Objective To evaluate the protective effects of soft-jujube kiwifruit extract on injured vascular endothelial cells(VEC).Methods Injured vascular endothelial cells were induced by hydrogen peroxide(H2O2),and the effects of extracts of soft-jujube kiwifruit extract on cell proliferation were detected by tetramethylazothiazol blue colorimetric method,and the effects on release of endothelin(ET-1)and prostacyclin(PGI2)were detected by radioimmunoassay.Results Hydrogen peroxide has a damaging effect on vascular endothelial cells.The extracts of soft-jujube kiwifruit extract can counteract proliferation of VEC and can inhibit LDH leakage,increase PGI2 release,and reduce ET-1 release in H2O2-induced vascular endothelial cell.Conclusion Soft-jujube kiwifruit extract possesses the Protective effects on injured vascular endothelial cells.
