秦皮药材HPLC指纹图谱研究 被引量:3

摘要 目的采用高效液相色谱法建立中药药材秦皮的指纹图谱,为秦皮药材科学的质量控制提供参考依据。方法采用Inertsil C18色谱柱(250 mm×4.6 mm,5μm),以体积分数0.1%甲酸水-甲醇溶液为流动相进行梯度洗脱,流速为0.8 mL·min^-1,检测波长为254 nm,柱温30℃;建立秦皮药材指纹图谱,运用中国药典委员会"中药色谱指纹图谱相似度评价系统软件"对10批秦皮药材进行相似度评价,并对其中四个色谱峰进行指认,对10批不同来源秦皮药材进行聚类分析。结果10批秦皮药材指纹图谱中标定了18个共有色谱峰,相似度值均在0.9以上。结论该法所建立的指纹图谱方法精密、重复性好、稳定性强,对提高秦皮药材的整体质量控制提供参考依据。 Objective To establish the HPLC fingerprint of Cortex Fraxini,one of the Chinese herbal medicine,which provides a reference for quality control of Cortex Fraxini scientifically.Methods The separation was performed on a Inertsil C18 column(250 mm×4.6 mm,5μm)with water(containing 0.1%formic acid)-methanol as the mobile phase by gradient elution at the flow rate of 0.8 mL·min^-1.The detection wavelength was 254 nm and the column temperature was 30℃.Fingerprints of Cortex Fraxini were established and the similarity of 10 batches of Cortex Fraxini was evaluated by using"System for Similarity Evaluation of Chromatographic Fingerprints of Traditional Chinese Medicines"of the Chinese Pharmacopoeia Commission.Results The fingerprint chromatogram had 18 common peaks,the four of the peaks were identified and the similarities of 10 batches of Cortex Fraxini were all above 0.9.Conclusion The established method for HPLC fingerprint of Cortex Fraxini is precise,reproducible and stable,which provides a reference basis for improving the overall quality evaluation of Cortex Fraxini.
