摘要
目的优化检测方法,探究可得然胶凝胶强度数据稳定,相对标准偏差小的检测方法。方法以可得然胶为材料,对比制胶方法,针对取样量、加热温度、冰水浴冷却时间、平行测定次数在单因素试验的基础上,根据正交试验对各因素条件进行优化,分析最优组合。结果确定检测方法为取样量为0.30~0.31 g,加入15 mL现制的脱气水,用微量取样药匙进行搅拌,使悬浊液分散均匀,沸水浴加热并维持其均匀度,水浴加热10 min后,取出在冰水浴中冷却50 min,做三次平行试验。结论该方法相对标准偏差小,数据稳定。且凝胶强度大于600 g·cm^(-2)的产品,可用直径0.4、0.5、0.6 cm中任一一款柱头进行测定。
Objective To optimize the detection method and explore the detection method with stable strength data and small relative standard deviation of Curdlan.Method Using Curdlan as the material,compare the glue making methods,optimize the sampling amount,heating temperature,ice water bath cooling time,and parallel measurement times based on single factor experiments,and analyze the optimal combination of each factor condition through orthogonal experiments.Result The determined detection method is to take a sample of 0.30-0.31 g,add 15 mL of freshly prepared degassed water,stir with a trace sampling spoon to evenly disperse the suspension,heat in a boiling water bath and maintain its uniformity,heat in a water bath for 10 minutes,then remove and cool in an ice water bath for 50 minutes,and conduct three parallel experiments.Conclusion This method has a small relative standard deviation and stable data.The column heads with diameters of 0.4 cm,0.5 cm and 0.6 cm for products with gel strength greater than 600 g·cm^(-2)can be used for determination.
