全蝎白术白头翁组方发酵前后萃取工艺及体外抗肿瘤活性研究

Study on Extraction Technology of Scorpio,Atractylodes Macrocephalae Rhizoma and Radix Pulsatillae Prescription Before and After Fermentation

摘要 为了研究全蝎白术白头翁组方(SAPZF)发酵前后最佳萃取部位,采用球孢白僵菌双向固体发酵技术制备全蝎白术白头翁组方发酵物(SAPZFFW),分别制取组方发酵前后的质量分数75%醇提液。采用系统萃取法,制得SAPZFFW、SAPZF的石油醚部、乙酸乙酯部、正丁醇部、水部干浸膏。通过MTT法检测SAPZFFW、SAPZF不同极性部位对乳腺癌细胞株MCF-7的抑制作用。结果表明:SAPZF有效部位的抗肿瘤能力由大到小依次为,石油醚部、正丁醇部、乙酸乙酯部;SAPZFFW有效部位抗肿瘤能力由大到小依次为,正丁醇部、石油醚部、乙酸乙酯部;SAPZFFW对乳腺癌细胞株MCF-7的抑制作用优于SAPZF,且正丁醇部效果较好。 In order tostudy the optimum extraction site of Scorpio,Atractylodes Macrocephalae Rhizoma and Radix Pulsatillae Prescription(SAPZF)before and after fermentation,SAPZFFW(fermentation product of SAPZF)was prepared by the two-way solid fermentation of Beauveria bassiana.The 75%alcohol extract of the SAPZFFW and SAPZF were subsequently obtained.Their dry extracts of petroleum ether part,ethyl acetate part,n-butanol part and water part were prepared by system extraction,respectively.Their inhibitions on breast cancer cell line MCF-7 were assayed by MTT for different polar sites.The anti-tumor ability of SAPZF effective site in order from large to small was determined as follows:petroleum ether part>n-butanol part>ethyl acetate part.That of SAPZFFW was determined as follows:n-butanol part>petroleum ether part>ethyl acetate part.The inhibitory effect of SAPZFFW on breast cancer cell line MCF-7 was better than that of SAPZF,and the effect of n-butylalcohol part was better.
