全蝎中多糖提取分离及其药理作用研究 被引量:12

摘要 在已有的全蝎酶解产物研究基础上,分离全蝎中的蛋白,提纯多糖,并研究全蝎中除去蛋白与未除蛋白多糖的抗炎作用。方法:用胰蛋白酶制备全蝎酶解液,乙醇沉出粗多糖,制备低纯多糖,Sevag法除去多糖中的蛋白质,测定全蝎中多糖的质量分数并进行抗炎研究。结论:未除蛋白组的多糖质量分数为42.12%,除去蛋白的多糖质量分数为87.27%,全蝎中多糖可抑制RAW264.7炎症反应,多糖蛋白对细胞炎症抑制作用不明显,除去蛋白后全蝎多糖对IL-6炎症因子的抑制效果不显著,对TNF-α炎症因子抑制效果显著,随药液浓度不断增大,抑制效果先上升后下降,当质量浓度为1.5mg·mL-1时,药液中TNF-α炎症因子质量浓度最低,说明该质量浓度下全蝎多糖的抗炎效果最为显著,减少了炎症因子的分泌,具有重要研究价值和市场前景。 On the basis of existing research on scorpion enzymatic hydrolysate,protein in Scorpio was separated,polysaccharide was purified,and anti-inflammatory effects of protein removed and non-protein removed polysaccharide in Scorpio were studied.Methods:Scorpio enzymolysis liquid was prepared by trypsin,crude polysaccharide was precipitated by ethanol,low pure polysaccharide was prepared,protein in polysaccharide was removed by sevag method,content of polysaccharide in Scorpio was determined and anti-inflammatory study was carried out.Conclusion:The polysaccharide content of the non-deproteinized group is 42.12%,the polysaccharide content of deproteinized group is 87.27%,the polysaccharide in Scorpio can inhibit RAW264.7 inflammatory reaction,the inhibitory effect of polysaccharide protein on cellular inflammation is not obvious,the inhibitory effect of Scorpio polysaccharide on IL-6 inflammatory factor after deproteinization is not significant,the inhibitory effect on TNF-αinflammatory factor is significant,with the increasing concentration of liquid medicine,the inhibitory effect first rises and then falls.When the concentration is 1.5 mg mL-1,the concentration of TNF-αinflammatory factor in the liquid medicine is the lowest,which indicates that Scorpio polysaccharide has the most significant anti-inflammatory effect at this concentration,reduces the secretion of inflammatory factor,and has important research value and market prospect.
