结直肠癌组织长链非编码RNA AOC4P表达及临床意义探讨

Expression and clinical significance of long noncoding RNA AOC4P in colorectal cancer

目的检测LncRNA AOC4P在结直肠癌组织中的表达,分析其与临床病理特征的相关性,为寻找结直肠癌肿瘤标志物及药物作用靶点提供参考。方法选取2018-01-01-2020-12-31泰安市中心医院收治68例结直肠癌患者癌组织及其癌旁组织标本为研究对象,采用实时荧光定量PCR检测结直肠癌组织以及癌旁组织LncRNA AOC4P表达,分析其与结直肠癌临床病理特征及预后相关性。结果实时荧光定量PCR结果显示,结直肠癌组织中LncRNA AOC4P表达量(0.308±0.126)低于癌旁组织,差异有统计学意义,t=5.180,P<0.001;Ⅰ期和Ⅱ期结直肠癌组织中LncRNA AOC4P表达量(0.391±0.129)高于Ⅲ期和Ⅳ期结直肠癌组织(0.247±0.081),差异有统计学意义,t=5.269,P<0.001;伴有远处转移结直肠癌组织中LncRNA AOC4P表达量(0.231±0.079)低于不伴有远处转移结直肠癌组织(0.359±0.126),差异有统计学意义,t=5.162,P<0.001。Kaplan-Meier生存曲线结果显示,结直肠癌组织中较高表达LncRNA AOC4P患者和较低表达LncRNA AOC4P患者相比,在相同时间节点生存率高,差异有统计学意义,对数秩检验,P=0.010。Cox回归分析表明,LncRNA AOC4P低表达是结直肠癌患者预后生存的独立危险因素,95%CI为0~0.1,P<0.05。结论LncRNA AOC4P在结直肠癌组织中表达减少,与临床病理特征显著相关,是患者预后生存的独立因素。

Objective To detect the expression of LncRNA AOC4P in colorectal cancer tissue,and then explore the relationship with clinical characteristics,in order to provide a theoretic foundation for the clinical research of colorectal cancer markers and related drug targets.Methods The expression of LncRNA AOC4P was detected in colorectal cancer tissues and adjacent normal tissues selected from 68 patients with colorectal cancer in Taian Central Hospital from 2018-01-01 to 2020-12-31 by real-time fluorescent quantitative PCR.Then the correlation analysis between lncRNA AOC4P expression and clinical features or survival time of patients suffered from colorectal cancer was done.Results The results of real-time fluorescent quantitative PCR showed that the expression of lncRNA AOC4P in colorectal cancer tissues(0.308±0.126)was significantly lower than that in normal tissues adjacent to the cancer(t=5.180,P<0.001).The results of real-time fluorescent quantitative PCR showed that the expression of lncRNA AOC4P inⅠandⅡcolorectal cancer(0.391±0.129)was significantly higher than that inⅢandⅣcolorectal cancer(0.247±0.081)(t=5.269,P<0.001).The results of real-time fluorescent quantitative PCR showed that the expression of lncRNA AOC4P with distant metastasis(0.231±0.079)was significantly lower than that without metastasis(0.359±0.126)(t=5.162,P<0.001).The results of Kaplan-Meier survival curve showed that patients with high expression of LncRNA AOC4P had a longer survival time than those with low expression of LncRNA AOC4P(log-rank test,P=0.010).The Cox regression showed that LncRNA AOC4P is an independent prognostic factor for overall survial in patients with colorectal cancer(95CI was 0-0.31,P<0.05).Conclusion The expression of LncRNA AOC4P is decreased in colorectal cancer tissues,which is related to clinical characteristics of colorectal cancer.It is an independent prognostic factor for overall survial in patients with colorectal cancer.
