摘要 通过高通量测序技术研究黑果腺肋花楸酒对D-半乳糖所致急性肝损伤小鼠肠道菌群的影响。饲喂昆明小鼠36只,以平均体重分为3组,分别是空白对照组(KB)12只、模型对照组(MX)12只、实验组(HQ)12只。全部小鼠适应饲喂一周后分组,其中KB组继续正常饲喂不作其它处理;MX组每天注射D-半乳糖(500 mg/kg),持续到实验结束;HQ组第一周与MX组相同,之后每天灌胃黑果腺肋花楸酒(2 mL/kg)至实验结束。共计饲喂5周,将小鼠处死前刺激排便取样进行16S r RNA的V3-V4区高通量测序,探究小鼠肠道菌群的变化。结果表明:MX组与KB组相比,菌群多样性、亲缘关系、物种多样性和丰度值均有很大差异,HQ组均与KB组相似;属水平分析,主势菌群含量最多的10种肠道菌总量,KB组、MX组和HQ组分别为35.2%、24.33%和36.36%。其中,拟杆菌属和乳杆菌属的菌群总量,MX组较KB组减少,而HQ组较MX组有所增加。表明黑果腺肋花楸酒对肝损伤小鼠的肠道菌群具一定的改善和修复作用。 In this experiment,high-throughput sequencing technology was used to study the effects of black chokeberry wine on the intestinal flora of mice with acute liver injury induced by D-galactose. Thirty six Kunming mice were raised for this experiment and divided into 3 groups with equivalent average body weight: 12 in the healthy control group(KB),12 in the injury model control group(MX),and 12 in the treated injury model group(HQ). Mice were adapted to their feeding regimen for one week prior to dividing them into groups. The KB group continued to be fed normally without other treatment. The MX and HQ groups were injected with D-galactose(500 mg/kg)daily until the end of the experiment. The HQ group was administered black chokeberry wine(2 mL/kg)every day until the end of the experiment. After feeding for a total of 5 weeks,the mice were assessed for increased defecation,then sacrificed for high-throughput sequencing of the V3-V4 region of the prokaryotic 16 S rRNA to explore changes in the intestinal flora of the mice. Compared with the KB group,the MX group had greater differences in flora diversity,genetic relationships,species diversity,and abundance values. The HQ group was similar to the KB group. According to the analysis of the genus level,the total biomasses comprised by the 10 most prevalent intestinal bacteria were 35.2%,24.33% and 36.36% in the KB,MX,and HQ groups,respectively. Among these,the total biomass of Bacteroides and Lactobacillus was reduced in the MX group compared with the KB group,while they were increased in the HQ group relative to the MX group,indicated that black chokeberry wine can improve and repair the intestinal flora of mice with liver injury.
