蛋白质组学在动物源中药研究中的应用

Application of proteomics in animal-derived traditional Chinese medicines

摘要 动物源中药是传统中药的重要组成部分,因其成分复杂不易分离分析,导致其活性成分和机制研究进度缓慢,也进一步增加了评价动物源药材的品种质量以及保障临床安全用药的难度。随着多维色谱、质谱、生物信息学等技术的飞速发展,蛋白质组学逐渐成为动物源中药研究的重要工具。蛋白质组学从整体水平对药材本身或药物处理前后的动物或细胞模型的总蛋白进行定性定量分析,在获得中药指纹图谱、挖掘潜在活性成分以及定位药物靶点和通路方面具有显著优势。本文主要介绍了蛋白质组学技术及其在动物源中药药材品种和质量鉴定、药用资源开发和可持续发展、药效物质基础及作用机制研究等方面的应用,为动物源中药的深入研究提供新的研究思路和方法。 Animal-derived medicines(AdM)are important parts of traditional Chinese medicine(TCM).The compositions of AdM are complex and the active ingredients are not easily analyzed,which makes the study of the mechanism of AdM proceeds slowly,and difficult to assess the quality of AdM and guarantee safety in clinical use.With the rapid development of multidimensional chromatography,mass spectrometry,bioinformatics and other technologies,proteomics has gradually become a powerful tool for the AdM.Qualitative and quantitative proteomics allows the analysis of total proteins extracted from animal or cellular samples after drug treatment at a holistic level,and has significant advantages in obtaining the fingerprint of TCM,discovering the potential active ingredients and clarifying the drug targets and pathways.This paper presents the main process of proteomics technology and its application in the identification and quality analysis of varieties of AdM,the maintenance of sustainable development and the utilization of Chinese medicine resources,and the mechanism of effective substances from AdM,providing new ideas and research methods based on proteomics for the in-deep studies of animal-derived traditional Chinese medicine.
