焦山楂炮制工艺及其改善胃动力障碍小鼠症状的研究

摘要 目的优化焦山楂炮制工艺,并探讨其对胃动力障碍小鼠症状的改善作用。方法采用热分析技术,研究山楂总黄酮、多糖等不同有效成分的热解特性,得到焦山楂的最佳炮制温度范围;以炒制时间、炒制温度为考察因素,以有机酸含量为指标结合响应曲面法实验优化焦山楂炮制工艺;并通过阿托品建立胃动力障碍小鼠模型,对焦山楂炮制工艺对胃动力障碍小鼠症状的改善作用进行对比研究。结果热分析实验综合分析得出焦山楂最佳炮制温度范围为216.0-257.5℃;响应面法优化得出焦山楂最优炮制工艺为炒制温度246℃,炒制时间8.05 min;传统焦山楂组与新法焦山楂组均可显著提高小鼠小肠推进率(P<0.05),而对胃残留率无显著影响(P>0.05);新法焦山楂组血清中胆囊收缩素(CCK)和胃泌素(GAS)的含量显著升高(P<0.05),且新法焦山楂组的效果要好于传统焦山楂组。结论热分析技术和响应面法并用优化得到的焦山楂炮制工艺具有可行性,且对胃动力障碍小鼠的症状具有改善作用,表明该法可为改进中药炮制工艺,合理量化炮制过程中火力火候参数提供借鉴。 Objective To optimize the processing technology of Hawthorn and to investigate its improvement effect on the symptom of gastric motility disorder mice.Methods The hawthorn flavones,polysaccharide and other different pyrolysis characteristics of effective ingredients,and get the best processing temperature range of coke hawthorn with thermal analysis technique,Taking the organic acid content in coke hawthorn as the index,two factors of stir-frying temperature and stir-frying time was chosen to optimize the processing technology of coke hawthorn by response surface methodology.The mouse model of gastric motility disorder was established by atropine,and the improvement of the symptoms of gastric motility disorder mice by focusing on coke hawthorn processing technology was compared.Results The optimum processing temperature range of coke hawthorn was 216.0-257.5℃.The response surface methodology was used to optimize the optimal processing process of coke hawthorn as processing temperature 246℃and processing time 8.05 min.The intestinal propulsion rate of mice in both the traditional coke hawthorn group and the new coke Hawthorn group was significantly increased(P<0.05),but the gastric residual rate was not significantly affected.The contents of CCK and GAS in serum of new coke hawthorn group were significantly increased(P<0.05),and the effect of new coke hawthorn group was better than that of traditional coke hawthorn group.Conclusion Thermal analysis technology and response surface methodology combined with the optimized processing technology of coke hawthorn is feasible,and it can improve the symptoms of mice with gastric motility disorder,indicating that the method can provide reference for improving the processing technology of Traditional Chinese medicine and reasonably quantifying the thermal temperature parameters in the process of processing.
