摘要
目的运用网络药理学方法分析预测血府逐瘀汤治疗失眠的作用机制。方法借助TCMSP、BATMAN-TCM数据库筛选血府逐瘀汤的有效活性成分及作用靶点;Genecards、OMIM获得失眠的疾病靶点,Cytoscape软件构建药物-活性成分-作用靶点网络并分析;STRING及Cytoscape构建PPI网络;利用Bioconductor对作用靶点进行GO功能和KEGG通路富集分析,预测其作用机制。结果血府逐瘀汤包含227个中药活性成分和303个靶点,作用于失眠的有110个活性成分和52个靶点。其中关键成分有槲皮素、7-甲氧基-2-甲基异黄酮、豆甾醇、山奈酚、β-谷甾醇,核心靶点为ALB、APP、IL-6、FOS。GO功能和KEGG通路富集分析结果显示血府逐瘀汤主要涉及神经递质水平的调节、细胞因子受体结合等生物过程,通过作用于糖尿病并发症的AGE-RAGE信号通路、IL-17信号通路、神经活性配体-受体相互作用等多条通路发挥治疗失眠的作用。结论本研究初步揭示了血府逐瘀汤治疗失眠的活性成分、作用靶点及通路,提示血府逐瘀汤具有多成分、多靶点、多途径的协同作用特点,为进一步深入研究其作用机制提供了重要依据。
Objective To analyze and predict the mechanism of Xuefu Zhuyu Decoction in treating insomnia by using network pharmacology.Methods TCMSP and BATMAN-TCM databases were used to screen the effective active ingredients and action targets of Xuefu Zhuyu Decoction;Genecards and OMIM were used to obtain the disease targets of insomnia,Cytoscape software was used to construct the drug-active ingredient-action target network and analyze them;STRING and Cytoscape were used to construct the PPI network;Bioconductor was used to analyze the GO function and KEGG pathway enrichment of the action targets and predict their mechanism of action.Results Xuefu Zhuyu Decoction contained 227 active ingredients of traditional Chinese medicine and 303 targets,acting on insomnia with 110 active ingredients and 52 targets.The key components were quercetin,7-methoxy-2-methyl isoflavone,stigmasterol,kaempferol,β-sitosterol.The core targets were ALB,APP,IL-6 and FOS.The results of the GO function and KEGG pathway enrichment analysis showed that Xuefu Zhuyu Decoction mainly involves the regulation of neurotransmitter levels,cytokine receptor binding and other biological processes,and exerts its therapeutic effect on insomnia by acting on multiple pathways such as AGE-RAGE signaling pathway,IL-17 signaling pathway,and neuroactive ligand-receptor interactions in diabetic complications.Conclusion This study has initially revealed the active ingredients,targets and pathways of Xuefu Zhuyu Decoction in treating insomnia,suggesting that Xuefu Zhuyu Decoction has the characteristics of multi-component,multi-target,and multi-path synergistic effects,which provides an important basis for further study of its mechanism.
