P2X7受体在择时电针调整小鼠自发活动近日节律中的影响研究

Study of the Effect of P2X7 Receptors in Timing Electroacupuncture to Adjust the Recent Rhythm of Spontaneous Activity in Rat

摘要 目的观察择时电针对野生型(WT)小鼠和P2X7基因敲除型(P2X7 KO)小鼠自发活动的影响,探讨P2X7受体在择时电针调整自发活动近日节律中的作用。方法WT小鼠36只,P2X7 KO小鼠36只,两类小鼠各自分为CT8、CT16两个时间点处理,每个时间点又分为空白组(Con组)、假穴组(SA组)、电针组(EA组),共计12组,每组6只。采用Achoff导引方法,电针组于暗置第11天电针"百会"穴和"长强"穴,假穴组于暗置第11天电针腹部非经非穴点,空白组不做处理,运用Clocklab、Matlab分析电针对小鼠自发活动近日节律的影响及相位转移值的大小。结果(1)在CT8时,WT电针组小鼠自发活动相位前置(P<0.01),P2X7 KO电针组小鼠自发活动相位前置(P<0.05或P<0.01);(2)在CT16时,WT电针组小鼠自发活动相位后置(P<0.05或P<0.01),P2X7 KO电针组小鼠自发活动相位后置(P<0.05);(3)在CT8时电针组中,与P2X7 KO小鼠比较,WT小鼠自发活动相位前置明显(P<0.05)。结论P2X7可能参与了CT8时电针对自发活动昼夜节律的影响,这可能是择时电针治疗近日节律紊乱的科学基础之一。 Objective To explore the role of P2 X7 receptors in the adjustment of spontaneous activity in recent rhythms by timing electroacupuncture in wild-type(WT)and P2 X7 knockout(P2 X7 KO)rats.Methods 36 WT rats and 36 P2 X7 KO rats were divided into CT8 and CT16 for treatment.Each time is divided into blank group(Con group),sham acupoint group(SA group),and electroacupuncture group(EA group).There are 12 groups in total,with 6 animals in each group.Applied the methods of Achoff Guides,the rats in the EA group were given acupuncture at Baihui(DU20)and Changqiang(DU1)points on the 11 th day in the dark,the rats in the SA group were given acupuncture at sham points on the 11 th day in the dark,and the blank group is not processed.Use Clocklab and Matlab to analyze the influence of electroacupuncture on spontaneous activities of rats and the magnitude of phase shift value.Results(1)At CT8,the phase of spontaneous activity of rats in the WT EA group was preceded(P<0.01),and the phase of spontaneous activity of rats in the P2 X7 KO EA group was preceded(P<0.05 or P<0.01).(2)At CT16,the phase of spontaneous activity of rats in the WT EA group was delayed(P<0.05 or P<0.01),and the phase of spontaneous activity of rats in the P2 X7 KO EA group was delayed(P<0.05).(3)In the EA group at CT8,compared with P2 X7 KO rats,the phase of spontaneous activity of WT rats was significantly preceded(P<0.05).Conclusion P2 X7 may be involved in the effect of CT8 on the circadian rhythm of spontaneous activity,which may be one of the scientific bases for the treatment of perihelion rhythm disorders by chronotropic electroacupuncture.
