藏药余甘子主要成分含量测定研究 被引量:11

摘要 目的建立高效液相色谱法(High Performance Liquid Chromatography,HPLC)同时测定藏药余甘子药材中主要成分没食子酸、安石榴苷B、没食子酸甲酯、老鹳草素、柯里拉京、诃子林鞣酸、诃黎勒酸和鞣花酸含量的方法,为余甘子药材质量控制提供依据。方法采用Diamonsil C18色谱柱(250×4.6 mm,5μm);流动相:甲醇-0.2%冰醋酸水;流速:1 mL·min-1;柱温:30℃;检测波长:270 nm。结果余甘子药材中主要成分没食子酸、安石榴苷B、没食子酸甲酯、老鹳草素、柯里拉京、诃子林鞣酸、诃黎勒酸和鞣花酸分别在0.094-1.87,0.10-2.51,0.047-1.49,0.12-3.84,0.28-8.80,0.055-1.75,0.042-1.34,0.0090-2.88μg线性范围内线性关系良好,平均回收率分别为:100.15,102.04,101.20,99.94,99.75,100.69,100.30,100.30%。结论该方法简便、准确、稳定,可同时测定余甘子药材主要成分的含量,为余甘子的质量控制提供依据。 Objective To establish a HPLC method for the simultaneous determination of gallic acid,punicalagin A,punicalagin B,methyl gallate,geraniin,corilagin,chebulinic acid,chebulagic acid,and ellagic acid in the Tibetan medicine Phyllanthus emblica L.and to provide basis for quality control of Phyllanthus emblica L.Method Mobile phase:A(methanol);B(H2 O:CH3 COOH,100:0.2,v/v).Column:DiamansilTM C18(250×4.6 mm,5μm),maintained at 30℃with flow rate of 1.0 mL.min-1.The detection wavelength was set at 270 nm for acquiring chromatograms.The injection volume was 20μL.Result The linear ranges of gallic acid,punicalagin A,punicalagin B,methyl gallate,geraniin,corilagin,chebulinic acid,chebulagic acid,and ellagic acid were 0.094-1.87,0.058-1.39,0.10-2.51,0.047-1.49,0.12-3.84,0.28-8.80,0.055-1.75,0.042-1.34,and 0.0090-2.88μg.The average recovery rates of gallic acid,punicalagin A,punicalagin B,methyl gallate,geraniin,corilagin,chebulinic acid,chebulagic acid,and ellagic acid were:100.15,102.04,101.20,99.94,99.75,100.69,100.30,100.30%.And RSDs were 0.63,3.76,3.37,1.08,1.53,1.22,0.98,1.98%.Conclusion This method is simple,accurate and stable,can simultaneously determin the contents of gallic acid,punicalagin A,punicalagin B.methyl gallate,geraniin,corilagin,chebulinic acid,chebulagic acid,and ellagic acid and can provide a basis for quality control of Phyllanthus emblica L.
