两种岭南湿热证小鼠模型肠道菌群动态变化的研究 被引量:21

摘要 目的基于两种岭南湿热证小鼠模型肠道菌群动态变化规律,寻求具有模型评价潜力的特征菌属。方法随机将52只BALB/c小鼠分为A1正常组、A2正常环境+肠产毒性大肠埃希氏菌(ETEC)组、A3正常环境+脂多糖(LPS)组、B1湿热环境组、B2湿热环境+ETEC组、B3湿热环境+LPS组。B1、B2、B3组予湿热环境、高脂高糖饮食,造模第20天,A2、B2组予ETEC,A3、B3组予LPS。观察一般情况,收集9次粪便,查结肠病理,测菌群、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6),采用SPSS 26.0软件统计。结果B2、B3组出现类似临床湿热证表现,结肠病理提示炎症,与A1组相比TNF-α、IL-6升高(P<0.05),模型成立前大肠杆菌属和拟杆菌属、成立时肠球菌属呈特征性增多(P<0.05),B1组"湿阻证模型"以肠球菌属增多为主,后期"热"证表现时梭菌属增多(P<0.05)。结论大肠杆菌属、拟杆菌属、肠球菌属在岭南湿热证模型客观评价指标的发掘上具有广阔前景,肠球菌属、梭菌属可能分别是"湿""、热"的特征菌属。 Objective To search for characteristic bacterial genus with potential in model evaluation by exploring the dynamic changes of intestinal flora in two kinds of mice models of Lingnan damp-heat syndrome.Methods Fifty-two BALB/c mice were randomly divided into A1 normal control group,A2 normal environment+Enterotoxigenic Escherichia coli(ETEC)group,A3 normal environment+Lipopolysaccharides(LPS)group,B1 hygrothermal environment group,B2 hygrothermal environment+ETEC group,and B3 hygrothermal environment+LPS group.Mice in group B1、B2 and B3 were lived in hygrothermal environment,and were fed high fat and high sucrose diet.On the 20 th day of modeling,group A2 and B2 were given ETEC,while group A3 and B3 were given LPS.We observed the general conditions and collected stool samples of mice for nine times.Then we checked the colonic pathology and detected the flora as well as the levels of TNF-α、IL-6.SPSS26.0 was used for statistics finally.Results Group B2 and B3 showed similar manifestations of damp-heat syndrome in clinic.Colonic pathology of them suggested inflammation,and their levels of TNF-αand IL-6 increased compared with group A1(P<0.05).Before the establishment of the model,the quantities of Escherichia coli and Bacteroides expanded,while Enterococcus had large growth when the models were established in group B2 and B3(P<0.05).In group B1,which was regarded as"the model of damp retention syndrome",Enterococcus increased mainly,while Clostridium grew in the later stage of"heat"syndrome(P<0.05).Conclusion Escherichia coli,Bacteroides,and Enterococcus have broad prospects in the exploration of objective evaluation indexes for the model of Lingnan damp-heat syndrome.Enterococcus and Clostridium may be the characteristic bacterial genus of"damp"and"heat"syndrome respectively.
