基于网络药理学的忍冬藤抗类风湿关节炎作用机制研究 被引量:17

摘要 目的利用网络药理学相关技术和方法,阐释忍冬藤抗类风湿关节炎的作用机制。方法通过检索数据库和文献,收集整理忍冬藤的化学成分、化学成分靶点和类风湿关节炎相关靶点,利用实体语法系统构建忍冬藤化学成分、化学成分靶点和类风湿关节炎相关靶点的生物网络,通过网络分析,辨识忍冬藤抗类风湿关节炎的有效成分群,阐释其作用机制。结果本研究通过网络拓扑结构分析得到有效成分31个、关键靶点13个,通过富集分析,得到忍冬藤抗类风湿关节炎的作用机制为:氮化合物代谢过程的正向调控、细胞生物合成过程的正向调控、转录的正调控、基因表达的正调控、靶向输送、调节脂质分解代谢过程、细胞分解代谢过程的正调控、聚合酶II启动子转录的正调控、细胞凋亡的正向调控、细胞死亡的正调控等。结论本研究较为系统地解析了忍冬藤治疗类风湿关节炎的作用机制,为临床治疗类风湿关节炎新药的研发和类风湿关节炎病因病机的研究奠定了基础。 Objective Network pharmacology related techniques and methods were used to elucidate the mechanism of action of Caulis Lonicerae against rheumatoid arthritis.Method Databases and literatures were searched to sort out the chemical components,chemical composition targets and targets associated with rheumatoid arthritis of Caulis Lonicerae.A biological network of chemical components,targets of chemical components and targets of rheumatoid arthritis was constructed by entity grammar system.Through network analysis,the effective components of Caulis Lonicerae against rheumatoid arthritis were identified and its mechanism was explained.Results 31 active components and 13 key targets were obtained by CytoNCA analysis.The action mechanisms of Caulis Lonicerae against rheumatoid arthritis were obtained by enrichment analysis:Positive regulation of nitrogen compound metabolic process,positive regulation of transcription,positive regulation of gene expression,protein targeting,regulation of lipid catabolic process,positive regulation of cellular catabolic process,positive regulation of transcription from RNA polymerase II promoter,positive regulation of apoptosis,positive regulation of cell death,etc.Conclusion This paper systematically analyzed the action mechanism of Caulis Lonicerae in the treatment of rheumatoid arthritis,lay a foundation for clinical treatment of rheumatoid arthritis,development of new drugs and the research of etiology and pathogenesis of rheumatoid arthritis.
