昆仑雪菊通过NOS/NO途径改善高脂饮食诱导低氧环境大鼠海马组织的损伤 被引量:3

Kunlun Chrysanthemum Ameliorate the Hippocampal Damage Induced by High Fat Diet in Rats Exposed to Hypoxia by NOS/NO Pathway

摘要 目的研究高脂饮食对低氧环境大鼠海马组织的影响及昆仑雪菊干预效果与机制。方法将40只雄性SD大鼠随机分为4组,分别为低氧对照(H)组、低氧高脂饮食(HH)组、昆仑雪菊干预低氧(XJH)组、昆仑雪菊干预低氧高脂饮食(XJHH)组。进行造模与昆仑雪菊提取液干预,实验周期为一周。光学显微镜观察海马病理变化,TBA比色法检测海马组织丙二醛含量,WST-1法检测超氧化物歧化酶活力,硝酸还原酶法检测硝酸盐/亚硝酸盐(NOx)水平、荧光实时定量RT-PCR检测海马NOS mRNA水平。结果与H组比,HH组SOD升高(P<0.05),MDA降低(P<0.05),iNOS mRNA升高(P<0.05),XJH组NO、MDA均升高(P<0.05),eNOS mRNA水平降低(P<0.05),nNOS mRNA升高(P<0.05);与HH组比,XJHH组MDA升高(P<0.05),nNOS mRNA升高(P<0.05)。结论①低氧环境可对大鼠海马产生损伤,集中在CA1区,高脂饮食加重此反应,原因为高脂饮食上调iNOS mRNA表达。②昆仑雪菊可减轻高脂饮食诱导的低氧环境大鼠海马的病理损伤,机制可能与上调nNOS mRNA表达介导的保护作用有关。 Objective We studied the effects of high-fat diet on the hippocampal tissue of rats in hypoxia environment and the intervention effects of kunlun chrysanthemum.Methods 40 male rats were randomly divided into:hypoxia(H)group,hypoxia and high-fat diet(HH)group,hypoxia and kunlun chrysanthemum(XJH)group,hypoxia and high-fat diet and kunlun chrysanthemum(XJHH)group.Mold making and extract intervention were carried out.The experiment lasted one week.The hippocampal morphological changes were observed by optical microscope.Malondialdehyde content was detected by TBA colorimetric method,superoxide dismutase activity was detected by WST-1 method,nitrate/nitrite(NOx)level was detected by nitrate reductase method,and the level of hippocampal NOS mRNA was detected by RT-PCR.Results Compared with the H group,the HH group showed increased SOD(P<0.05),decreased MDA(P<0.05),and significantly increased iNOS mRNA(P<0.05),the XJH group showed significantly increased NO and MDA(P<0.05),decreased eNOS mRNA(P<0.05),and increased nNOS mRNA(P<0.05).Compared with HH group,the nNOS mRNA and MDA in XJHH group were increased(P<0.05).Conclusion①Hypoxia may cause damage in hippocampus,mainly in the CA1 region,and high-fat diet can aggravate this damage caused by increasing iNOS mRNA.②Kunlun chrysanthemum can ameliorate the damage in hippocampus of rats under hypoxia and high fat diet,which may be achieved by adjusting the expression of nNOS mRNA and then achieving the effects of hippocampal protection.
