维库溴铵对人非小细胞肺癌A549细胞株增殖和转移能力影响分析

摘要 目的探讨维库溴铵对非小细胞肺癌A549细胞株增殖能力和转移能力的影响作用。方法2017年1月—2019年2月,将该院44例非小细胞肺癌患者随机等分为两组,每组各22例,分离获取全部入选患者的非小细胞肺癌A549细胞株样本,在体外环境中实施细胞培养处理,为参照组样本运用标准化杜氏高糖培养基实施培养,为研究组运用标准化杜氏高糖培养基联合20.00μg/mL维库溴铵溶液实施培养,对比两组的PI3K/AKT通路相关蛋白表达量指标测算值、A549细胞增殖倍数指标测算值,以及A549细胞凋亡率指标测算值。结果研究组的PI3K表达量指标测算值(0.77±0.04)μg/mL低于参照组(0.90±0.05)μg/mL,差异有统计学意义(t=9.523,P<0.05)。研究组的p-AKT/AKT表达量指标测算值(0.72±0.10)μg/mL低于参照组(0.86±0.08)μg/mL,差异有统计学意义(t=5.128,P<0.05)。研究组的p-mTOR/mTOR表达量指标测算值(0.74±0.09)μg/mL低于参照组(0.80±0.12)μg/mL,差异有统计学意义(t=1.876,P<0.05)。研究组的HIF-1α表达量指标测算值(0.73±0.05)μg/mL低于参照组(0.84±0.08)μg/mL,差异有统计学意义(t=5.469,P<0.05)。研究组的A549细胞增殖倍数指标测算值(3.31±0.37)倍低于参照组(5.50±0.41)倍,差异有统计学意义(t=18.600,P<0.05)。研究组的A549细胞凋亡率指标测算值(19.5±2.2)%高于参照组(4.8±1.0)%,差异有统计学意义(t=28.531,P<0.05)。结论维库溴铵对非小细胞肺癌A549细胞株的增殖能力与转移能力有影响。 Objective To investigate the effect of vecuronium on the proliferation and metastasis ability of non-small cell lung cancer A549 cells.Methods From January 2017 to February 2019,44 patients with non-small cell lung cancer were randomLy divided into two groups,22 cases in each group.All non-small cell lung cancer A549 cell lines were selected.The cell culture treatment was car-ried out in an in vitro environment,and the reference group samples were cultured using a standardized Duss high glucose medium,and the research group was cultured using a standardized Duss high glucose medium in combination with 20.00μg/mL vecuronium bromide solution,and the two groups were compared.The PI3 K/AKT pathway-related protein expression index,the A549 cell proliferation factor index,and the A549 cell apoptosis rate index.Results The PI3 K expression index(0.77±0.04)μg/mL in the study group was lower than the reference group(0.90±0.05)μg/mL,and there was a difference between the groups the difference was statistically significant(t=9.523,P<0.05).The calculated value of p-AKT/AKT expression index(0.72±0.10)μg/mL in the study group was lower than that in the reference group(0.86±0.08)μg/mL,and there was a difference between the groups the difference was statistically significant(t=5.128,P<0.05).The p-mTOR/mTOR expression index of the study group(0.74±0.09)μg/mL was lower than the reference group(0.80±0.12)μg/mL,and there was a difference between the groups the difference was statistically significant(t=1.876,P<0.05).The HIF-1αexpression index of the study group(0.73±0.05)μg/mL was lower than the reference group(0.84±0.08)μg/mL,and there were differences between the groups the difference was statistically significant(t=5.469,P<0.05).In the study group,the A549 cell proliferation factor index(3.31±0.37)times was lower than the reference group(5.50±0.41)times,and there were differences between the groups the difference was statistically significant(t=18.600,P<0.05).The A549 cell apoptosis rate index(19.5±2.2)%was higher in the study group than in the reference group(4.8±1.0)%,and there was a difference between the groups the difference was statistically significant(t=28.531,P<0.05).Conclusion Vecuronium has an effect on the proliferation and metastasis ability of non-small cell lung cancer A549 cells.
