冬凌草配方颗粒超高效液相色谱指纹图谱及多指标成分含量测定方法的建立

Establishment of UPLC fingerprint and determination of multi-index components of Rabdosiae Rubescentis Herba formula granules

目的建立冬凌草配方颗粒的超高效液相色谱(UPLC)法指纹图谱,且对冬凌草配方颗粒中多成分含量进行测定。方法采用沃特世(Waters)ACQUITY UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8μm),以乙腈、0.1%甲酸水溶液为流动相进行梯度洗脱,流速0.4 mL·min,检测波长350 nm,柱温为30℃,进样量为3μL。结果建立了10批冬凌草配方颗粒的指纹图谱,得到8个共有峰,并指认了4个成分峰,分别是咖啡酸、阿魏酸、迷迭香酸、胡麻素,相似度在0.999~1.000之间。4种指标成分在各自浓度范围内呈良好的线性关系(R~2=0.9997~1.0000),平均加样回收率为90.27%~110.64%,RSD为0.07%~2.16%。结论该法重复性好,稳定可行,符合指纹图谱的分析要求,同时测定4种指标成分的含量作为其质量控制方法。

Objective To establish an ultra high performance liquid chromatography(UPLC)fingerprint of Rabdosiae Rubescentis Herba formula granules and determine the multi-component contents as a quality control method.Methods Waters ACQUITY UPLC HSS T3 column(2.1 mm×100 mm,1.8μm)was used with acetonitrile and 0.1%formic acid aqueous solution as mobile phase for gradient elution with a flow rate of 0.4 mL·min.The detection wavelength was 350 nm,the column temperature was 30℃,and the injection volume was 3μL.Results The fingerprints of 10 batches of Rabdosiae Rubescentis Herba formula granules were established,8 common peaks were obtained,and 4 component peaks were identified,namely caffeic acid,ferulic acid,rosmarinic acid and pedalitin,with a similarity of 0.999~1.000.The four components showed a good linear relationship in their respective concentration range(R~2=0.9997~1.0000).The average recoveries were between 90.27%and 110.64%with RSD of 0.07%~2.16%.Conclusion This method has good repeatability,stability and feasibility,meets the analysis requirements of fingerprint,and can determine four components at the same time,which provides a reference basis for establishing the quality control standard of Rabdosiae Rubescentis Herba formula granules.
