摘要
本文报告羊成骨蛋白(sBMP)对L929纤维母细胞培养的DNA合成和细胞分裂作用结果。所加sBMP浓度为1~10μg/ml时,L929纤维母细胞培养72小时后,细胞计数明显高于对照组,可达对照组的1.5~1.8倍,其细胞DNA合成过程中~3H胸腺嘧啶摄入量显著增加,可为对照组的145~164%。sBMP浓度5μg/ml和10μg/ml组中,细胞连续培养10天,未见其形态发生异常变化。实验结果表明sBMP对纤维母细胞DNA的合成和细胞的有丝分裂有显著的促进作用,但无促分化的作用。
The effect of sheep bone morphogenetic protein(sBMP),a substance extracted from demineralized bone matrix,was observed in L929 fibroblast culture.At concentrations of 1-10μg/ml,sBMP stimulated the incorporation of~3H-thymidine into DNA of the cultured cells by 145-164%in 72 hours,and caused a marked increase of cell number to 1.5-1.6-fold.The optimal concentration of sBMP was 5μg/ml.No change in differentiation of the cultured cells occur red within ten-day incubation period.These results indicate that sBMP stimulates DNA synthesis and mitotic division in fibroblast culture but does not result in the differentiation of the cultured fibroblasts.
